Immunohistochemistry Procedure . Specimen Preparation: Formalin-fixed, paraffin-embedded tissues are suitable for use with this primary antibody. Deparaffinization: Deparaffinize slides using xylene or xylene alternative and graded alcohols. Antibody Dilution: If using the concentrate format of this product, dilute the antibody 1:50. The dilutions are estimates; actual results may differ because of variability in methods and protocols. Antigen Retrieval: Boil tissue section in 10mM citrate buffer, pH 6.0 for 10 min followed by cooling at room temperature for 20 min. Primary Antibody Incubation: Incubate for 30 minutes at room temperature. Slide Washing: Slides must be washed in between steps. Rinse slides with PBS/0.05% Tween. Ribosomal S6 kinase has two isoforms, termed S6K1 (S6Ka) and S6K2 (S6Kb). The S6K1 isoform has two splicing forms, which are the products of alternative start codon usage. The first splicing form, p70S6K or p70S6K1, has a predicted molecular mass of 56kDa and an apparent molecular mass (SDS-PAGE) of 70kDa. The second isoform, p85S6K or p85S6K1, contains 23 extra amino acid residues at the N terminus, resulting from the alternative start codon. It has a predicted molecular mass of 59kDa and an apparent molecular mass of 85kDa. Both, p70 and p85 S6K1 are activated by multiple S/T phosphorylations in response to various mitogenic stimuli and nutrients. This is accompanied by a significant mobility shift in the migration of both forms when analyzed by SDS-PAGE electrophoresis.

This gene encodes a member of the ribosomal S6 kinase family of serine/threonine kinases. The encoded protein responds to mTOR (mammalian target of rapamycin) signaling to promote protein synthesis, cell growth, and cell proliferation. Activity of this gene has been associated with human cancer. Alternatively spliced transcript variants have been observed. The use of alternative translation start sites results in isoforms with longer or shorter N-termini which may differ in their subcellular localizations. There are two pseudogenes for this gene on chromosome 17. [provided by RefSeq, Jan 2013]
