storage stability :
Store at 2-8 degree C. Do not freeze. The user must validate any other storage conditions. When properly stored, the reagent is stable to the date indicated on the label. Do not use the reagent beyond the expiration date. There are no definitive signs to indicate instability of this product; therefore, positive and negative controls should be tested simultaneously with unknown specimens. If unexpected results are observed which cannot be explained by variations in laboratory procedures and a problem with the reagent is suspected, contact Technical Support.
tested application :
Immunohistochemistry (IHC) Paraffin
app notes :
Immunohistochemistry Procedure . Specimen Preparation: Formalin-fixed, paraffin-embedded tissues are suitable for use with this primary antibody. Deparaffinization: Deparaffinize slides using xylene or xylene alternative and graded alcohols. Antibody Dilutions: If using the concentrate format of this product, dilute the antibody 1:50 prior to use. The dilutions are estimates; actual results may differ because of variability in methods and protocols. Antigen Retrieval: Boil tissue section in 10mM Citrate buffer, pH 6.0 for 10-20 min followed by cooling at room temperature for 20 min. Primary Antibody: Incubate for 30 minutes at room temperature. Slide Washing: Slides must be washed in between steps. Rinse slides with PBS/0.05% Tween. Visualization: Detect the antibody as instructed by the instructions provided with the visualization system. Immunohistochemistry Dilution: 1:50. IHC Positive Control: Seminoma
other info2 :
Immunogen: Synthetic peptide corresponding to internal region of human OCT-3/OCT-4 protein. Control Tissue: Seminoma. Cellular Localization: Nucleus. Entrez Gene Code: 5460. Pathway: Stem Cell
products description :
OCT-3 (also known as OCT-4) is a mammalian POU transcription factor expressed by early embryo cells and germ cells. OCT-3/4 is essential for the identity of the pluripotential founder cell population in the mammalian embryo. A critical amount of OCT-3/4 is required to sustain stem cell renewal, and up or down regulation induce divergent developmental programs.

ncbi summary :
This gene encodes a transcription factor containing a POU homeodomain that plays a key role in embryonic development and stem cell pluripotency. Aberrant expression of this gene in adult tissues is associated with tumorigenesis. This gene can participate in a translocation with the Ewing's sarcoma gene on chromosome 21, which also leads to tumor formation. Alternative splicing, as well as usage of alternative AUG and non-AUG translation initiation codons, results in multiple isoforms. One of the AUG start codons is polymorphic in human populations. Related pseudogenes have been identified on chromosomes 1, 3, 8, 10, and 12. [provided by RefSeq, Oct 2013]
uniprot summary :
Oct4: Transcription factor that binds to the octamer motif (5 -ATTTGCAT-3 ). Forms a trimeric complex with SOX2 on DNA and controls the expression of a number of genes involved in embryonic development such as YES1, FGF4, UTF1 and ZFP206. Critical for early embryogenesis and for embryonic stem cell pluripotency. Interacts with UBE2I and ZSCAN10. Interacts with PKM2. Interacts with WWP2. Transcriptional activity is positively regulated by PKM2. Expressed in developing brain. Highest levels found in specific cell layers of the cortex, the olfactory bulb, the hippocampus and the cerebellum. Low levels of expression in adult tissues. Belongs to the POU transcription factor family. Class- 5 subfamily. 2 isoforms of the human protein are produced by alternative splicing. Protein type: Transcription factor; DNA-binding. Chromosomal Location of Human Ortholog: 6p21.31. Cellular Component: nucleoplasm; transcription factor complex; cytoplasm; nucleus; cytosol. Molecular Function: protein binding; miRNA binding; DNA binding; sequence-specific DNA binding; ubiquitin protein ligase binding; transcription factor activity; transcription factor binding. Biological Process: transcription from RNA polymerase II promoter; blastocyst development; anatomical structure morphogenesis; regulation of transcription, DNA-dependent; regulation of asymmetric cell division; regulation of gene expression; somatic stem cell maintenance; endodermal cell fate specification; response to wounding; positive regulation of transcription from RNA polymerase II promoter; mRNA transcription from RNA polymerase II promoter
