Immunohistochemistry Procedure . Specimen Preparation: Formalin-fixed, paraffin-embedded tissues are suitable for use with this primary antibody. Deparaffinization: Deparaffinize slides using xylene or xylene alternative and graded alcohols. Antibody Dilution: If using the concentrate format of this product, dilute the antibody 1:25. The dilutions are estimates; actual results may differ because of variability in methods and protocols. Antigen Retrieval: Boil tissue section in 10mM citrate buffer, pH 6.0 for 10 min followed by cooling at room temperature for 20 min. Primary Antibody Incubation: Incubate for 30 minutes at room temperature. Slide Washing: Slides must be washed in between steps. Rinse slides with PBS/0.05% Tween. Visualization: Detect the antibody as instructed by the instructions provided with the visualization system. Immunohistochemistry Dilution: 1:25. IHC Positive Control: Breast Carcinoma

The serine threonine protein kinase encoded by the AKT1 gene is catalytically inactive in serum starved primary and immortalized fibroblasts. AKT1 and the related AKT2 are activated by platelet derived growth factor. The activation is rapid and specific. In the developing nervous system AKT is a critical mediator of growth factor induced neuronal survival. Survival factors can suppress apoptosis in a transcription independent manner by activating the serine/threonine kinase AKT1, which then phosphorylates and inactivates components of the apoptotic machinery. The antibody labels activated AKT-1 that is phosphorylated at 473 serine residue. Multiple alternatively spliced transcript variants have been found for this gene (referenced from entrez gene).

Survival factors can suppress apoptosis in a transcription-independent manner by activating the serine/threonine kinase AKT1, which then phosphorylates and inactivates components of the apoptotic machinery.
