Deparaffinization: Deparaffinize slides using xylene or xylene alternative and graded alcohols. Antibody Dilution: If using the concentrate format of this product, dilute the antibody 1:50. The dilutions are estimates; actual results may differ because of variability in methods and protocols. Antigen Retrieval: Boil tissue sections in 10mM Citrate buffer, pH 6.0 for 10 min followed by cooling at RT for 20 min. Primary Antibody Incubation: Incubate for 10 minutes at RT. PMP-22 (Peripheral Myelin Protein) is a glycoprotein expressed in the compact myolin of the peripheral nervous system. In the peripheral nervous system, PMP-22 is produced by myelinating Schwann cells and is co-expressed with the genes for myelin basic protein (MBP) and protein P0 during nerve development and regeneration. Alterations in the level of this protein cause several genetic human diseases. If the protein is duplicated, patients develop Charcot-Marie-Tooth disease. If one copy of the gene is deleted, they suffer from the inherited tendency to pressure palsies.
