Immunohistochemistry Procedure . Specimen Preparation: Formalin-fixed, paraffin-embedded tissues are suitable for use with this primary antibody. Deparaffinization: Deparaffinize slides using xylene or xylene alternative and graded alcohols. Antibody Dilution: If using the concentrate format of this product, dilute the antibody 1:50. The dilutions are estimates; actual results may differ because of variability in methods and protocols. Antigen Retrieval: Boil tissue section in 10mM citrate buffer, pH 6.0 for 10 min followed by cooling at room temperature for 20 min. Primary Antibody Incubation: Incubate for 10 minutes at room temperature. Slide Washing: Slides must be washed in between steps. Rinse slides with PBS/0.05% Tween. Adults express two forms of glutamate decarboxylase (GAD), GAD65 and GAD67, which are encoded by different independently regulated genes. Both are expressed in most GABA containing neurons. GAD65 appears to be targeted to the membrane and nerve endings; while GAD67 is more widely distributed throughout the cell. GAD65 preferentially synthesizes GABA for vesicular release and GAD67 preferentially synthesizes cytoplasmic GABA. GAD65 is predominantly expressed in pancreatic beta cells and auto-antibodies to GAD65 which are strongly associated with insulin-dependent diabetes mellitus (IMDM).

This gene encodes one of several forms of glutamic acid decarboxylase, identified as a major autoantigen in insulin-dependent diabetes. The enzyme encoded is responsible for catalyzing the production of gamma-aminobutyric acid from L-glutamic acid. A pathogenic role for this enzyme has been identified in the human pancreas since it has been identified as an autoantibody and an autoreactive T cell target in insulin-dependent diabetes. This gene may also play a role in the stiff man syndrome. Alternative splicing results in multiple transcript variants that encode the same protein. [provided by RefSeq, Oct 2008]
