Store at 2-8 degree C. Do not freeze. The user must validate any other storage conditions. When properly stored, the reagent is stable to the date indicated on the label. Do not use the reagent beyond the expiration date. There are no definitive signs to indicate instability of this product; therefore, positive and negative controls should be tested simultaneously with unknown specimens. If unexpected results are observed which cannot be explained by variations in laboratory procedures and a problem with the reagent is suspected, contact Technical Support.

Immunohistochemistry (IHC) Paraffin
app notes :
Immunohistochemistry Procedure . Specimen Preparation: Formalin-fixed, paraffin-embedded tissues are suitable for use with this primary antibody. Deparaffinization: Deparaffinize slides using xylene or xylene alternative and graded alcohols. Antibody Dilution: If using the concentrate format of this product, dilute the antibody 1:50. The dilutions are estimates; actual results may differ because of variability in methods and protocols. Antigen Retrieval: Boil tissue section in 10mM citrate buffer, pH 6.0 for 10 min followed by cooling at room temperature for 20 min. Primary Antibody Incubation: Incubate for 30 minutes at room temperature. Slide Washing: Slides must be washed in between steps. Rinse slides with PBS/0.05% Tween. Visualization: Detect the antibody as instructed by the instructions provided with the visualization system. Immunohistochemistry Dilution: Ready To Use. IHC Positive Control: Placenta
other info2 :
Immunogen: Recombinant protein encoding aa 251-478 of human MMP-3. Control Tissue: Placenta. Cellular Localization: Cytoplasm. Entrez Gene Code: 4314. Pathway: Proteases and Protease Regulators
products description :
Matrix metalloproteinases (MMP) are proteolytic enzymes capable of degrading connective tissue components. Imbalanced secretion of certain MMP or disturbances in the differential control of MMP by tissue inhibitor of MMPs (TIMPs) is implicated in the invasive potential of malignant tumors. Stromelysin (MMP3/Transin) degrades several matrix components including the core proteins of proteoglycans, laminin and non-helical regions of collagens.

ncbi summary :
Proteins of the matrix metalloproteinase (MMP) family are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. Most MMP's are secreted as inactive proproteins which are activated when cleaved by extracellular proteinases. This gene encodes an enzyme which degrades fibronectin, laminin, collagens III, IV, IX, and X, and cartilage proteoglycans. The enzyme is thought to be involved in wound repair, progression of atherosclerosis, and tumor initiation. The gene is part of a cluster of MMP genes which localize to chromosome 11q22.3. [provided by RefSeq, Jul 2008]
uniprot summary :
MMP3: Can degrade fibronectin, laminin, gelatins of type I, III, IV, and V; collagens III, IV, X, and IX, and cartilage proteoglycans. Activates procollagenase. Defects in MMP3 are the cause of susceptibility to coronary heart disease type 6 (CHDS6). A multifactorial disease characterized by an imbalance between myocardial functional requirements and the capacity of the coronary vessels to supply sufficient blood flow. Decreased capacity of the coronary vessels is often associated with thickening and loss of elasticity of the coronary arteries. A polymorphism in the MMP3 promoter region is associated with the risk of coronary heart disease and myocardial infarction, due to lower MMP3 proteolytic activity and higher extracellular matrix deposition in atherosclerotic lesions. Belongs to the peptidase M10A family. Protein type: Secreted, signal peptide; Motility/polarity/chemotaxis; EC 3.4.24.17; Protease; Secreted. Chromosomal Location of Human Ortholog: 11q22.3. Cellular Component: proteinaceous extracellular matrix; extracellular space; mitochondrion; extracellular region; cytosol. Molecular Function: zinc ion binding; metalloendopeptidase activity; endopeptidase activity; calcium ion binding. Biological Process: collagen catabolic process; positive regulation of protein oligomerization; extracellular matrix disassembly; extracellular matrix organization and biogenesis; negative regulation of protein kinase B signaling cascade; protein catabolic process; proteolysis; regulation of cell migration. Disease: Coronary Heart Disease, Susceptibility To, 6
