Principle of the assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to TNF. Standards or samples are then added to the appropriate microtiter plate wells with a HRP-conjugated antibody preparation specific for TNF and incubated. Then substrate solution is added to each well. Only those wells that contain TNF and HRP-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of TNF in the samples is then determined by comparing the O.D. of the samples to the standard curve.

This gene encodes a multifunctional proinflammatory cytokine that belongs to the tumor necrosis factor (TNF) superfamily. This cytokine is mainly secreted by macrophages. It can bind to, and thus functions through its receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. This cytokine is involved in the regulation of a wide spectrum of biological processes including cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. This cytokine has been implicated in a variety of diseases, including autoimmune diseases, insulin resistance, and cancer.
