This assay has high sensitivity and excellent specificity for detection of Guinea pig TNF. No significant cross-reactivity or interference between Guinea pig TNF and analogues was observed.

Store at 2-8 degree C.

Samples: Serum, plasma and other biological fluids. Detection Range: 120 pg/mL - 2100 pg/mL. Sensitivity: The limit of detection of Guinea pig TNF defined as the analyte concentration resulting in an absorbance significantly higher than that of the dilution medium (mean plus 2 standard deviations) was determined to be 56 pg/mL (mean of 6 independent assays).

Intra-assay Precision: Intra-assay Precision (Precision within an assay) Three samples of known concentration were tested twenty times on one plate to assess intra-assay precision. Intra-Assay: CV<8%. Inter-assay Precision: Inter-assay Precision (Precision between assays) Three samples of known concentration were tested in forty separate assays to assess inter-assay precision. Principle of the assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to TNF. Standards or samples are then added to the appropriate microtiter plate wells with a HRP-conjugated antibody preparation specific for TNF and incubated. Then substrate solution is added to each well. Only those wells that contain TNF and HRP-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of TNF in the samples is then determined by comparing the O.D. of the samples to the standard curve.

This gene encodes a multifunctional proinflammatory cytokine that belongs to the tumor necrosis factor (TNF) superfamily. This cytokine is mainly secreted by macrophages. It can bind to, and thus functions through its receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. This cytokine is involved in the regulation of a wide spectrum of biological processes including cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. This cytokine has been implicated in a variety of diseases, including autoimmune diseases, insulin resistance, and cancer.
