This assay has high sensitivity and excellent specificity for detection of Guinea pig IFN-gamma. No significant cross-reactivity or interference between Guinea pig IFN-gamma and analogues was observed.

Store at 2-8 degree C.

Samples: Serum, plasma and other biological fluids. Detection Range: 15.6 pg/mL - 1000 pg/mL. The standard curve concentrations used for the ELISA's were 1000 pg/mL, 500 pg/mL, 250 pg/mL, 125 pg/mL, 62.5 pg/mL, 31.25 pg/mL, 15.6 pg/mL, 0 pg/mL. Sensitivity: The limit of detection of Guinea pig IFN-gamma defined as the analyte concentration resulting in an absorbance significantly higher than that of the dilution medium (mean plus 2 standard deviations) was determined to be 3.9 pg/mL (mean of 6 independent assays). Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitative IFN-gamma in Guinea pig serum, plasma. An antibody specific for IFN-gamma has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any IFN-gamma present is bound by the immobilized antibody. After removing any unbound substances, a biotin - conjugated antibody specific for IFN-gamma is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of IFN-gamma bound in the initial step. The color development is stopped and the intensity of the color is measured.
