This assay has high sensitivity and excellent specificity for detection of Human F2RL3. No significant cross-reactivity or interference between Human F2RL3 and analogues was observed.

Samples: Serum, plasma, tissue homogenates and other biological fluids. Assay Type: Sandwich. Detection Range: 15.6 pg/mL - 1000 pg/mL. The standard curve concentrations used for the ELISA's were 1000 pg/mL, 500 pg/mL, 250 pg/mL, 125 pg/mL, 62.5 pg/mL, 31.25 pg/mL, 15.6 pg/mL, 0 pg/mL . Sensitivity: The limit of detection of Human F2RL3 defined as the analyte concentration resulting in an absorbance significantly higher than that of the dilution medium (mean plus 2 standard deviations) was determined to be 12.8 pg/mL (mean of 6 independent assays).

Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitative F2RL3 in Human serum, plasma, tissue homogenates. An antibody specific for F2RL3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any F2RL3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin - conjugated antibody specific for F2RL3 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of F2RL3 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Coagulation factor II (thrombin) receptor-like 3 (F2RL3) is a member of the large family of 7-transmembrane-region receptors that couple to guanosine-nucleotide-binding proteins. F2RL3 is also a member of the protease-activated receptor family. F2RL3 is activated by proteolytic cleavage of its extracellular amino terminus. The new amino terminus functions as a tethered ligand and activates the receptor. F2RL3 is activated by thrombin and trypsin. [provided by RefSeq, Jul 2008]
