Intended Uses: The kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of FcgR3A in human serum, plasma, tissue homogenates, cell lysates and other biological fluids. Principle of the Assay: The microplate provided in this kit has been pre-coated with an antibody specific to FcgR3A. Standards or samples are then added to the appropriate microplate wells with a biotin-conjugated antibody specific to FcgR3A. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain FcgR3A, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of FcgR3A in the samples is then determined by comparing the O.D. of the samples to the standard curve.

This gene encodes a receptor for the Fc portion of immunoglobulin G, and it is involved in the removal of antigen-antibody complexes from the circulation, as well as other other antibody-dependent responses. This gene (FCGR3A) is highly similar to another nearby gene (FCGR3B) located on chromosome 1. The receptor encoded by this gene is expressed on natural killer (NK) cells as an integral membrane glycoprotein anchored through a transmembrane peptide, whereas FCGR3B is expressed on polymorphonuclear neutrophils (PMN) where the receptor is anchored through a phosphatidylinositol (PI) linkage. Mutations in this gene have been linked to susceptibility to recurrent viral infections, susceptibility to systemic lupus erythematosus, and alloimmune neonatal neutropenia.
