Principle of the Assay: This experiment use double-sandwich elisa technique and the ELISA Kit provided is typical. The pre-coated antibody is Rat NMDA monoclonal antibody and the detecting antibody is polyclonal antibody with biotin labeled. Samples and biotin labeling antibody are added into ELISA plate wells and washed out with PBS or TBS. Then Avidin-peroxidase conjugates are added to ELISA wells in order; Use TMB substrate for coloring after reactant thoroughly washed out by PBS or TBS. TMB turns into blue in peroxidase catalytic and finally turns into yellow under the action of acid. The color depth and the testing factors in samples are positively correlated.

A similar gene in mice encodes a protein that plays a critical role in spatial learning and memory by regulating the function of synaptic N-methyl-D-aspartic acid receptor complexes in the hippocampus. NETO1: a transmembrane protein and component of the N-methyl-D-aspartate receptor (NMDAR) complex. Required to maintain the normal level of NMDAR subunits at synapses. The NMDAR complex is a major excitatory ligand-gated ion channel in the central nervous system and a major mediator of synaptic plasticity. Important in the development and/or maintenance of neuronal circuitry. Neto1-deficient mice have a dramatic decrease in the number of NMDA receptors at synapses, impairing learning and synaptic plasticity. Three isoforms of the human protein are produced by alternative splicing. Isoforms 1 and 2 are retina-specific whereas isoform 3 is found in retina as well as at lower levels in adult and fetal brain. Protein type: Membrane protein, integral.
