products description :
Priciple of the assay: This experiment use double-sandwich elisa technique and the ELISA Kit provided is typical. The pre-coated antibody is human OBSCN monoclonal antibody and the detecting antibody is polyclonal antibody with biotin labeled. Samples and biotin labeling antibody are added into ELISA plate wells and washed out with PBS or TBS. Then Avidin-peroxidase conjugates are added to ELISA wells in order; Use TMB substrate for coloring after reactant thoroughly washed out by PBS or TBS. TMB turns into blue in peroxidase catalytic and finally turns into yellow under the action of acid. The color depth and the testing factors in samples are positively correlated.

ncbi summary :
The obscurin gene spans more than 150 kb, contains over 80 exons and encodes a protein of approximately 720 kDa. The encoded protein contains 68 Ig domains, 2 fibronectin domains, 1 calcium/calmodulin-binding domain, 1 RhoGEF domain with an associated PH domain, and 2 serine-threonine kinase domains. This protein belongs to the family of giant sacromeric signaling proteins that includes titin and nebulin, and may have a role in the organization of myofibrils during assembly and may mediate interactions between the sarcoplasmic reticulum and myofibrils. Alternatively spliced transcript variants encoding different isoforms have been identified. [provided by RefSeq, Jul 2008]
uniprot summary :
OBSCN: a Ca(2+)/calmodulin-dependent protein kinase of the Trio family. A giant protein with one Rho GEF domain, one PH domain and two kinase domains. Expressed in skeletal and cardiac muscle, concentrating around Z-disks and M-lines of striated muscle. Binds ankyrin 1 interacts with sarcomeric myosin. May play a critical role in its ability to assemble into A bands in striated muscle. Protein type: EC 2.7.11.1; Kinase, protein; Protein kinase, CAMK; Protein kinase, Ser/Thr (non-receptor); CAMK group; Trio family. Chromosomal Location of Human Ortholog: 1q42.13. Cellular Component: myofibril; M band; cytosol; Z disc. Molecular Function: calmodulin binding; protein serine/threonine kinase activity; Rho guanyl-nucleotide exchange factor activity; protein binding; structural constituent of muscle; metal ion binding; titin binding; ankyrin binding; ATP binding. Biological Process: regulation of small GTPase mediated signal transduction; nerve growth factor receptor signaling pathway; positive regulation of apoptosis; small GTPase mediated signal transduction; multicellular organismal development; sarcomere organization; protein amino acid phosphorylation
