products description :
Intended Uses: This ELISA kit applies to the invitro quantitative determination of HumanKRT1concentrations in serum, plasma and other biological fluids. Principle of the Assay: This ELISA kit uses Sandwich-ELISA as the method. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to KRT1. Standards or samples are added to the appropriate micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for KRT1and Avidin-Horseradish Peroxidase (HRP) conjugate is added to each micro plate well successively and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain KRT1, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The OD value is proportional to the concentration of KRT1.You can calculate the concentration of KRT1in the samples by comparing the OD of the samples to the standard curve.

ncbi summary :
The protein encoded by this gene is a member of the keratin gene family. The type II cytokeratins consist of basic or neutral proteins which are arranged in pairs of heterotypic keratin chains coexpressed during differentiation of simple and stratified epithelial tissues. This type II cytokeratin is specifically expressed in the spinous and granular layers of the epidermis with family member KRT10 and mutations in these genes have been associated with bullous congenital ichthyosiform erythroderma. The type II cytokeratins are clustered in a region of chromosome 12q12-q13. [provided by RefSeq, Jul 2008]
uniprot summary :
K1: a type II cytoskeletal keratin. The keratins are intermediate filament proteins responsible for the structural integrity of epithelial cells and are subdivided into cytokeratins and hair keratins. There are two types of cytoskeletal and microfibrillar keratin: type I (acidic; 40-55 kDa) [K9 to K20] and type II (neutral to basic; 56-70 kDa) [K1 to K8]. Both a basic and an acidic keratin are required for filament assembly. Generally associates with K10. Protein type: Cytoskeletal. Chromosomal Location of Human Ortholog: 12q13.13. Cellular Component: cytoskeleton; extracellular matrix; extracellular space; keratin filament; membrane; nucleus; plasma membrane. Molecular Function: carbohydrate binding; protein binding; receptor activity; structural molecule activity. Biological Process: complement activation, lectin pathway; fibrinolysis; negative regulation of inflammatory response; regulation of angiogenesis; response to oxidative stress; retinal homeostasis. Disease: Epidermolytic Hyperkeratosis; Ichthyosis Hystrix, Curth-macklin Type; Ichthyosis, Cyclic, With Epidermolytic Hyperkeratosis; Keratosis Palmoplantaris Striata Iii; Palmoplantar Keratoderma, Epidermolytic; Palmoplantar Keratoderma, Nonepidermolytic
