products description :
Intended Uses: This ELISA kit applies to the in vitro quantitative determination of Porcine OPN concentrations in serum, plasma and other biological fluids. Principle of the Assay This ELISA kit uses Sandwich-ELISA as the method. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to OPN. Standards or samples are added to the appropriate micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for OPN and Avidin-Horseradish Peroxidase (HRP) conjugate is added to each micro plate well successively and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain OPN, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The OD value is proportional to the concentration of OPN. You can calculate the concentration of OPN in the samples by comparing the OD of the samples to the standard curve.

ncbi summary :
The protein encoded by this gene is involved in the attachment of osteoclasts to the mineralized bone matrix. The encoded protein is secreted and binds hydroxyapatite with high affinity. The osteoclast vitronectin receptor is found in the cell membrane and may be involved in the binding to this protein. This protein is also a cytokine that upregulates expression of interferon-gamma and interleukin-12. Several transcript variants encoding different isoforms have been found for this gene. uniprot summary :
osteopontin: Binds tightly to hydroxyapatite. Appears to form an integral part of the mineralized matrix. Probably important to cell-matrix interaction. Belongs to the osteopontin family. 5 isoforms of the human protein are produced by alternative splicing. Protein type: Secreted; Secreted, signal peptide. Chromosomal Location of Human Ortholog: 4q22.1. Cellular Component: extracellular space; cell projection; perinuclear region of cytoplasm; apical part of cell; extracellular region. Molecular Function: protein binding; extracellular matrix binding; cytokine activity. Biological Process: osteoblast differentiation; neutrophil chemotaxis; extracellular matrix disassembly; extracellular matrix organization and biogenesis; response to vitamin D; biomineral formation; response to steroid hormone stimulus; negative regulation of collateral sprouting of intact axon in response to injury; positive regulation of bone resorption; decidualization; inflammatory response; cell adhesion; embryo implantation
