This kit recognizes natural and recombinant Rat PAI-1. No significant cross-reactivity or interference between Rat PAI-1 and analogues was observed.

Sensitivity: The minimum detectable dose of Rat PAI-1 is 0.188ng/mL (The sensitivity of this assay, or lowest detectable limit (LDL) was defined as the lowest protein concentration that could be differentiated from zero).

Intended Uses: This ELISA kit applies to the in vitro quantitative determination of Rat PAI-1 concentrations in serum, plasma and other biological fluids.

Principle of the assay: This ELISA kit uses Sandwich-ELISA as the method. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to PAI-1. Standards or samples are added to the appropriate micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for PAI-1 and Avidin-Horseradish Peroxidase (HRP) conjugate is added to each micro plate well successively and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain PAI-1, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of PAI-1. You can calculate the concentration of PAI-1 in the samples by comparing the OD of the samples to the standard curve.

This gene encodes a member of the serine proteinase inhibitor (serpin) superfamily. This member is the principal inhibitor of tissue plasminogen activator (tPA) and urokinase (uPA), and hence is an inhibitor of fibrinolysis. Defects in this gene are the cause of plasminogen activator inhibitor-1 deficiency (PAI-1 deficiency), and high concentrations of the gene product are associated with thrombophilia.
