Intended Uses: This ELISA kit applies to the in vitro quantitative determination of SheepANG II concentrations in serum, plasma and other biological fluids. Principle of the Assay: This ELISA kit uses Competitive-ELISA as the method. The microtiter plate provided in this kit has been pre-coated with ANG II. During the reaction,ANG II in the sample or standard competes with a fixed amount ofANG IIon the solid phase supporterfor sites on the Biotinylated Detection Ab specific toANG II. Excess conjugate and unbound sample or standard are washed from the plate, andAvidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Thena TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration ofANG II in the samples is then determined by comparing the OD of the samples to the standard curve.

In response to lowered blood pressure, the enzyme renin cleaves angiotensinogen to produce angiotensin-1 (angiotensin 1-10). Angiotensin-1 is a substrate of ACE (angiotensin converting enzyme) that removes a dipeptide to yield the physiologically active peptide angiotensin-2 (angiotensin 1- 8).
