products description :
Intended Uses: This ELISA kit applies to the in vitro quantitative determination of Bovine, Horse, Sheep E2 concentrations in serum and plasma samples. Please contact tech-support for other sample type detection. Principle of the Assay: This ELISA kit uses the Competitive-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with E2. During the reaction, E2 in the sample or standard competes with a fixed amount of E2 on the solid phase supporter for sites on the Biotinylated Detection Ab specific to E2. Excess conjugate and unbound sample or standard are washed from the plate, and Avidin conjugated to Horseradish Peroxidase (HRP) are added to each microplate well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of stop solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of E2 in the samples is then determined by comparing the OD of the samples to the standard curve!!Background/Introduction: Estradiol is a female sex hormone produced by the ovaries, adrenal gland and also the placenta during pregnancy. Estradiol is the most important hormone during a female's reproductive years, and is required for reproductive and sexual function as well as having an impact on the health of other organs and tissues. In females, estradiol acts primarily as a growth hormone for the reproductive organs including the vagina, the fallopian tubes, the endometrium and the cervical glands [1]. Estradiol also enhances growth of the womb's muscle layer, the myometrium. In addition, the hormone maintains oocytes and triggers a series of events that lead to ovulation. Though estradiol levels in men are much lower compared to those in women, estradiol has important roles in men as well. Aside from humans and other mammals, estradiol is also found in most vertebrates and crustaceans, insects, fish, and other animal species. Estradiol is produced especially within the follicles of the ovaries, but also in other tissues including the testicles, the adrenal glands, fat, liver, the breasts, and the brain. Estradiol is produced in the body from cholesterol through a series of reactions and intermediates [2].
