products description :
Intended Uses This ELISA kit applies to the in vitro quantitative determination of Human WARS concentrations in serum, plasma and other biological fluids. Principle of the Assay: This ELISA kit uses Sandwich-ELISA as the method. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to WARS. Standards or samples are added to the appropriate micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for WARS and Avidin-Horseradish Peroxidase (HRP) conjugate is added to each micro plate well successively and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain WARS, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The OD value is proportional to the concentration of WARS. You can calculate the concentration of WARS in the samples by comparing the OD of the samples to the standard curve.

ncbi summary :
Aminoacyl-tRNA synthetases catalyze the aminoacylation of tRNA by their cognate amino acid. Because of their central role in linking amino acids with nucleotide triplets contained in tRNAs, aminoacyl-tRNA synthetases are thought to be among the first proteins that appeared in evolution. Two forms of tryptophanyl-tRNA synthetase exist, a cytoplasmic form, named WARS, and a mitochondrial form, named WARS2. Tryptophanyl-tRNA synthetase (WARS) catalyzes the aminoacylation of tRNA(trp) with tryptophan and is induced by interferon. Tryptophanyl-tRNA synthetase belongs to the class I tRNA synthetase family. Four transcript variants encoding two different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]
uniprot summary :
WARS: Isoform 1, isoform 2 and T1-TrpRS have aminoacylation activity while T2-TrpRS lacks it. Isoform 2, T1-TrpRS and T2-TrpRS possess angiostatic activity whereas isoform 1 lacks it. T2-TrpRS inhibits fluid shear stress-activated responses of endothelial cells. Regulates ERK, Akt, and eNOS activation pathways that are associated with angiogenesis, cytoskeletal reorganization and shear stress-responsive gene expression. Belongs to the class-I aminoacyl-tRNA synthetase family. 2 isoforms of the human protein are produced by alternative splicing. Protein type: Amino Acid Metabolism - tryptophan; Translation; Ligase; EC 6.1.1.2. Chromosomal Location of Human Ortholog: 14q32.31. Cellular Component: cytoplasm; nucleus; cytosol. Molecular Function: protein binding; tryptophan-tRNA ligase activity; ATP binding. Biological Process: negative regulation of cell proliferation; tRNA aminoacylation for protein translation; tryptophanyl-tRNA aminoacylation; translation; gene expression; angiogenesis; regulation of angiogenesis
