specificity :
This assay has high sensitivity and excellent specificity for detection of Human VE-Cadherin. No significant cross-reactivity or interference between Human VE-Cadherin and analogues was observed. Note: Limited by current skills and knowledge, it is impossible for us to complete the cross- reactivity detection between Human VE-Cadherin and all the analogues, therefore, cross reaction may still exist.
storage stability :
Store at 4 degree C.

products description :
Description: The kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of VE-Cadherin in human serum, plasma and other biological fluids. Principle of the Assay: This ELISA kit uses Sandwich-ELISA as the method. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Human VE-Cadherin. Standards or samples are added to the appropriate micro ELISA plate wells and bound by the specific antibody. Then a biotinylated detection antibody specific for Human VE-Cadherin and Avidin-Horseradish Peroxidase (HRP) conjugate is added to each micro plate well successively and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Human VE-Cadherin, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The OD value is proportional to the concentration of Human VE-Cadherin. You can calculate the concentration of Human VE-Cadherin in the samples by comparing the OD of the samples to the standard curve.

ncbi summary :
This gene is a classical cadherin from the cadherin superfamily and is located in a six-cadherin cluster in a region on the long arm of chromosome 16 that is involved in loss of heterozygosity events in breast and prostate cancer. The encoded protein is a calcium-dependent cell-cell adhesion glycoprotein comprised of five extracellular cadherin repeats, a transmembrane region and a highly conserved cytoplasmic tail. Functioning as a classic cadherin by imparting to cells the ability to adhere in a homophilic manner, the protein may play an important role in endothelial cell biology through control of the cohesion and organization of the intercellular junctions. An alternative splice variant has been described but its full length sequence has not been determined. [provided by RefSeq, Jul 2008]
uniprot summary :
CDH5: Cadherins are calcium dependent cell adhesion proteins. They preferentially interact with themselves in a homophilic manner in connecting cells; cadherins may thus contribute to the sorting of heterogeneous cell types. This cadherin may play a important role in endothelial cell biology through control of the cohesion and organization of the intercellular junctions. It associates with alpha-catenin forming a link to the cytoskeleton. Acts in concert with KRIT1 to establish and maintain correct endothelial cell polarity and vascular lumen. These effects are mediated by recruitment and activation of the Par polarity complex and RAP1B. Required for activation of PRKCZ and for the localization of phosphorylated PRKCZ, PARD3, TIAM1 and RAP1B to the cell junction. Interacts via cadherin 5 domain with PTPRB. Interacts with TRPC4. Interacts with KRIT1. Endothelial tissues and brain. Protein type: Motility/polarity/chemotaxis; Cell adhesion; Membrane protein, integral. Chromosomal Location of Human Ortholog: 16q22.1. Cellular Component: adherens junction; tight junction; membrane; plasma membrane; integral to membrane; intercellular junction; cell junction. Molecular Function: protein binding; beta-catenin binding; calcium ion binding; protein phosphatase binding; receptor binding. Biological Process: intercellular junction assembly and maintenance; negative regulation of cell proliferation; homophilic cell adhesion; vascular endothelial growth factor receptor signaling pathway; blood vessel maturation
