CD59_HUMAN
specificity :
This assay has high sensitivity and excellent specificity for detection of Human CD59. No significant cross-reactivity or interference between Human CD59 and analogues was observed. Note: Limited by current skills and knowledge, it is impossible for us to complete the cross- reactivity detection between Human CD59 and all the analogues, therefore, cross reaction may still exist.
storage stability :
Store at 4 degree C.
other info1 :
Samples: Serum, Plasma, Biological Fluids. Assay Type: Sandwich. Detection Range: 31.25--2000pg/mL. Sensitivity: 18.75pg/mL
products description :
Description: The kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of CD59 in human serum, plasma and other biological fluids. Principle of the Assay: This ELISA kit uses Sandwich-ELISA as the method. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Human CD59. Standards or samples are added to the appropriate micro ELISA plate wells and bound by the specific antibody. Then a biotinylated detection antibody specific for Human CD59 and Avidin-Horseradish Peroxidase (HRP) conjugate is added to each micro plate well successively and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Human CD59, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The OD value is proportional to the concentration of Human CD59. You can calculate the concentration of Human CD59 in the samples by comparing the OD of the samples to the standard curve.

ncbi summary :
This gene encodes a cell surface glycoprotein that regulates complement-mediated cell lysis, and it is involved in lymphocyte signal transduction. This protein is a potent inhibitor of the complement membrane attack complex, whereby it binds complement C8 and/or C9 during the assembly of this complex, thereby inhibiting the incorporation of multiple copies of C9 into the complex, which is necessary for osmolytic pore formation. This protein also plays a role in signal transduction pathways in the activation of T cells. Mutations in this gene cause CD59 deficiency, a disease resulting in hemolytic anemia and thrombosis, and which causes cerebral infarction. Multiple alternatively spliced transcript variants, which encode the same protein, have been identified for this gene. [provided by RefSeq, Jul 2008]
uniprot summary :
CD59: Potent inhibitor of the complement membrane attack complex (MAC) action. Acts by binding to the C8 and/or C9 complements of the assembling MAC, thereby preventing incorporation of the multiple copies of C9 required for complete formation of the osmolytic pore. This inhibitor appears to be species-specific. Involved in signal transduction for T-cell activation complexed to a protein tyrosine kinase. Defects in CD59 are the cause of CD59 deficiency (CD59D). Protein type: Membrane protein, GPI anchor. Chromosomal Location of Human Ortholog: 11p13. Cellular Component: anchored to external side of plasma membrane; extracellular space; compact myelin; cell surface; focal adhesion; membrane; plasma membrane; extracellular region; vesicle; sarcolemma. Molecular Function: complement binding; protein binding. Biological Process: cell activation; cell surface receptor linked signal transduction; regulation of complement activation; negative regulation of activation of membrane attack complex; innate immune response; positive regulation of T cell proliferation; blood coagulation; negative regulation of apoptosis. Disease: Hemolytic Anemia, Cd59-mediated, With Or Without Immune-mediated Polyneuropathy
