products description :
Intended Uses: This ELISA kit applies to the in vitro quantitative determination of Rat TIMP-2 concentrations in serum, plasma and other biological fluids. Principle of the Assay: This ELISA kit uses Sandwich-ELISA as the method. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to TIMP-2. Standards or samples are added to the appropriate micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for TIMP-2 and Avidin-Horseradish Peroxidase (HRP) conjugate is added to each micro plate well successively and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain TIMP-2, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of TIMP-2. You can calculate the concentration of TIMP-2 in the samples by comparing the OD of the samples to the standard curve.

acts as an inhibitor of metallopeptidase activity; may play a role in tissue remodeling during spermatogenesis [RGD, Feb 2006]
uniprot summary :
TIMP2: Complexes with metalloproteinases (such as collagenases) and irreversibly inactivates them by binding to their catalytic zinc cofactor. Known to act on MMP-1, MMP-2, MMP-3, MMP-7, MMP-8, MMP-9, MMP-10, MMP-13, MMP-14, MMP-15, MMP-16 and MMP-19. Interacts (via the C-terminal) with MMP2 (via the C- terminal PEX domain); the interaction inhibits the MMP2 activity. Down-regulated by TGFB1. Belongs to the protease inhibitor I35 (TIMP) family. Protein type: Secreted, signal peptide; Motility/polarity/chemotaxis; Secreted; Extracellular matrix; Inhibitor; Cell development/differentiation. Cellular Component: extracellular space; proteinaceous extracellular matrix; neuron projection; growth cone; cell surface; cell soma; basement membrane; nucleus. Molecular Function: integrin binding; metalloendopeptidase inhibitor activity; protease binding; metal ion binding; enzyme activator activity. Biological Process: response to drug; negative regulation of proteolysis; positive regulation of catalytic activity; regulation of Rap protein signal transduction; negative regulation of metalloenzyme activity; central nervous system development; regulation of neuron differentiation; regulation of cAMP metabolic process; response to hormone stimulus; negative regulation of membrane protein ectodomain proteolysis; negative regulation of cell proliferation; positive regulation of MAPKKK cascade; response to cytokine stimulus; regulation of MAPKKK cascade; negative regulation of Ras protein signal transduction; spermatogenesis; negative regulation of mitotic cell cycle; positive regulation of adenylate cyclase activity; positive regulation of neuron differentiation; aging
