Usage: Immunohistochemistry: MBS241618 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for MBS241618 was determined to be 5 ug/ml.

Rinse slides in 1X TBST -1 minute @ RT. Apply alkaline phosphatase chromogen substrate - 30 minutes @ RT. Wash slides in distilled water - 1 minute @ RT. Dehydrate: . (This method should only be used if the chromogen substrate is alcohol insoluble (e.g. Vector Red, DAB) . Wash slides in 2 changes of 80% alcohol - 1 minute each @ RT. Wash slides in 2 changes of 95% alcohol - 1 minute each @ RT. Wash slides in 3 changes of 100% alcohol - 1 minute each @ RT. Wash slides in 3 changes of xylene - 1 minute each @ RT. Apply coverslip
products description :
NOS1 / nNOS belongs to the family of nitric oxide synthases, which synthesize nitric oxide from L-arginine. Nitric oxide is a reactive free radical, which acts as a biologic mediator in several processes, including neurotransmission, and antimicrobial and antitumoral activities. In the brain and peripheral nervous system, nitric oxide displays many properties of a neurotransmitter, and has been implicated in neurotoxicity associated with stroke and neurodegenerative diseases, neural regulation of smooth muscle, including peristalsis, and penile erection. This protein is ubiquitously expressed, with high level of expression in skeletal muscle. Multiple transcript variants that differ in the 5' UTR have been described for this gene but the full-length nature of these transcripts is not known. Additionally, alternatively spliced transcript variants encoding different isoforms (some testis-specific) have been found for this gene.
