CREB (cAMP response element binding protein1), a DNA binding protein, is a member of the leucine zipper family of transcription factors. CREB interaction with DNA requires phosphorylation at Ser 119 by activated cAMP protein kinase A. Once phosphorylated CREB binds to an 8 base enhancer know as the cAMP response element (CRE) containing a conserved core sequence TGACG. CREB also interacts with CBP (CREB binding protein) and RNA polymerase II when phosphorylated on Ser 133.

This protein binds as a homodimer to the cAMP-responsive element, an octameric palindrome. The protein is phosphorylated by several protein kinases, and induces transcription of genes in response to hormonal stimulation of the cAMP pathway.
