Target Species: Human. Immunogen Description: Synthetic peptide from human HSP90AA1 / Hsp90. Usage Summary: Immunohistochemistry: MBS240566 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for MBS240566 was determined to be 2.5 ug/ml. Summary: Hsp90 is a member of the heat shock protein 90 family that functions as a molecular chaperone and has ATPase activity. Hsp90 family proteins are highly conserved between isoforms and species. Several signal transduction pathways depend on Hsp90 function including erbB2, steroid hormone receptors (such as androgen, progesterone, glucocorticoid, and aryl-hydrocarbon), and hypoxia sensing (Hif1 alpha). Recent reports show that tumor cells are more sensitive to Hsp90 inhibition and that Hsp90 from tumor cells is more sensitive to small molecule inhibitors (eg 17AAG). The mechanism of this differential sensitivity of normal versus tumor Hsp90 is not known (although mutation has been ruled out). One possible mechanism may be differences in post-translational modification of tumor Hsp90. Hsp90 is a cytoplasmic protein that forms a homodimer in vivo, and interacts with TOM34, AHSA1, HDAC6 and SMYD3 .
other info2 :
Immunogen Type: Synthetic peptide. Immunogen: HSP90AA1 / Hsp90 antibody was raised against synthetic peptide from human HSP90AA1 / Hsp90. Antigen Modification: aa289-300
products description :
HSP90AA1 is a member of the group of proteins produced by cells to deal with stress such as elevated temperature. It is known to chaperone a key subset of signaling proteins and is necessary for malignant transformation. HSP90 is subject to an array of post translational modifications that affect its function including acetylation.

ncbi summary :
The protein encoded by this gene is an inducible molecular chaperone that functions as a homodimer. The encoded protein aids in the proper folding of specific target proteins by use of an ATPase activity that is modulated by co-chaperones. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jan 2012]
uniprot summary :
HSP90A: a molecular chaperone of the heat shock protein 90 family. Has ATPase activity. Known to interact with a wide variety of proteins including steroid hormone receptors, neuropeptide Y, FKBP51/54, and FKBP52. G protein-coupled receptor kinases are stabilized by interacting with HSP 90. Hsp70 and Hsp90 promote tau solubility and tau binding to microtubules, reducing insoluble tau phosphorylation of tau. Protein type: Heat shock protein; Chaperone. Chromosomal Location of Human Ortholog: 14q32.33. Cellular Component: nucleoplasm; mitochondrion; membrane; cytoplasm; plasma membrane; melanosome; extracellular region; nucleus; cytosol. Molecular Function: identical protein binding; protein binding; protein homodimerization activity; TPR domain binding; nitric-oxide synthase regulator activity; unfolded protein binding; ATPase activity; nucleotide binding; ATP binding. Biological Process: axon guidance; receptor-mediated endocytosis; positive regulation of nitric oxide biosynthetic process; organelle organization and biogenesis; signal transduction; nitric oxide metabolic process; protein import into mitochondrial outer membrane; response to unfolded protein; mitochondrial transport; innate immune response; protein refolding; mitotic cell cycle; regulation of nitric-oxide synthase activity; vascular endothelial growth factor receptor signaling pathway; chaperone-mediated protein complex assembly; G2/M transition of mitotic cell cycle
