ELISA: Recombinant human IL-7 may be used as a standard for ELISA applications with either anti-Human IL-7 (purified) or anti-Human IL-7 (biotin conjugate). Western Blot: Recombinant human IL-7 may be used as the positive control for western blotting applications with either anti-Human IL-7 (purified) or anti-Human IL-7 (biotin conjugate). ELISA: Minimum Dilution: 0.2; Maximum Dilution: 0.4 ng/well. Functional Assays: Minimum Dilution: 0.1ng/ml; Maximum Dilution: 10ng/ml. Reconstitution: Reconstitute with 0.1 ml distilled water. For extended storage the addition of 0.1% BSA is recommended.Care should be taken during reconstitution as the protein may appear as a film at the bottom of the vial. Recombinant human IL-7 is 17.4 kDa protein comprised of 153 amino acids corresponing to the ful-length mature sequence. IL-7 is a potent lymphoid cell growth factor acting on Pre-B, Pro-B and early T cells. In a dose dependent cell proliferation assay using 2E8 cells this product shows a specific activity of > 2 x 106 units/mg. Recombinant human IL-7 is recognized by Rat anti Human IL-7 antibody clones BVD10-40F6 and BVD10-11C10 in ELISA.

The protein encoded by this gene is a cytokine important for B and T cell development. This cytokine and the hepatocyte growth factor (HGF) form a heterodimer that functions as a pre-pro-B cell growth-stimulating factor. This cytokine is found to be a cofactor for V(D)J rearrangement of the T cell receptor beta (TCRB) during early T cell development. This cytokine can be produced locally by intestinal epithelial and epithelial goblet cells, and may serve as a regulatory factor for intestinal mucosal lymphocytes. Knockout studies in mice suggested that this cytokine plays an essential role in lymphoid cell survival. Alternative splicing results in multiple transcript variants encoding distinct isoforms. Additional splice variants have been described but their presence in normal tissues has not been confirmed.[provided by RefSeq, Dec 2010]
