Synthetic human Osteopontin also known as Bone sialoprotein 1, nephropontin, secreted phosphoprotein 1, urinary stone protein or uropontin is a peptide corresponding to amino acids 17-27 of native human osteopontin, itself a 314 amino acid (including a 16 amino acid N-terminal signal peptide) multifunctional glycoprotein highly expressed in bone and a number of cell types including macrophages, endothelial and smooth muscle cells (Mazzali et al. 2002). The N-terminal peptide sequence for synthetic osteopontin is present in all five isoforms of human osteopontin described (UniProt: P10451). Polymorphisms in the osteopontin gene appear to contribute to susceptibility to systemic lupus erythematosus (D'Alfonso et al. 2005) and elevation of serum osteopontin levels correlate with disease severity in these patients (Wong et al. 2005).

The protein encoded by this gene is involved in the attachment of osteoclasts to the mineralized bone matrix. The encoded protein is secreted and binds hydroxyapatite with high affinity. The osteoclast vitronectin receptor is found in the cell membrane and may be involved in the binding to this protein. This protein is also a cytokine that upregulates expression of interferon-gamma and interleukin-12. Several transcript variants encoding different isoforms have been found for this gene.
