specificity :
This item specifically recognises 49kDa MEK5, also known as MAP2K5, when phosphorylated at Ser311 and Thr315. MEK5 is a dual specificity serine/threonine protein kinase belonging to the MAP kinase kinase family, which acts as a key regulator in a mitogen-activated protein (MAP) kinase cascade, and hence growth factor-induced cellular stimulation. MEK5 exists in naturally occurring MEK5alpha and MEK5beta splice variants, the former of which is known to specifically activate/regulate MAPK7/ERK5/BMK1, whilst the latter may act as a suppressor of MAPK7 activation. Activation of MEK5 itself is regulated by upstream MAP3K3/MEKK3, and phosphorylation of MEK5 at both the Ser311 and Thr315 sites has been implicated in cell proliferation.
form :
Purified. Purified IgG - liquid
storage stability :
Store at 4 degree C or at -20 degree C if preferred. Storage in frost-free freezers is not recommended. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use. Shelf Life: 12 months from date of despatch.
tested application :
Western Blot (WB)*
app notes :
Western Blot: This item detects a band of approximately 49kDa in rat testes cell lysates. Western Blotting: Maximum Dilution: 1/1000; Application Note: For the detection of phosphoproteins, tyrosine phosphatase inhibitors such as 1mM Sodium Orthovanadate should be added to the sample buffer. Milk or other casein-based blocking solutions are not recommended as casein is a phosphoprotein and its use can result in high background.
other info1 :
Perservative Stabilisers: 0.09% Sodium Azide (NaN3). 0.01% Bovine Serum Albumin. 50% Glycerol
other info2 :
Immunogen: Synthetic phosphopeptide corresponding to an amino acid sequence within MEK5, which includes phoshorylated Ser311 and Thr315. Buffer Solution: 10mM Hepes pH7.5. Antiserum Preparation: Antisera to MEK5 were raised by repeated immunisations of rabbits with highly purified antigen. Purified IgG was prepared by affinity chromatography. Target Species: Rat
