reactivity :
Mouse (Expected from Sequence)
specificity :
This item recognises the p53 - upregulated modulator of apoptosis (PUMA), also known as Bcl-2 binding component 3. Bcl-2 binding component 3 is a pro-apoptotic member of the BCL-2 family. Bcl-2 binding component 3 is localised to the mitochondria, binds BCL-2 and induces cytochrome C release and apoptosis in response to p53.
form :
Purified. Purified IgG - liquid
concentration :
IgG concentration 1.0 mg/ml
storage stability :
Store at 4 degree C or at -20 degree C if preferred. This product should be stored undiluted. Storage in frost-free freezers is not recommended. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use. Shelf Life: 18 months from date of despatch.
tested application :
Immunohistology Paraffin*, Western Blot (WB)
app notes :
Western Blot: This item detects a band of approximately 23kDa in K562 cell lysates. Immunohistology - Paraffin: Maximum Dilution: 10 ug/ml; Application Note: This product requires antigen retrieval using heat treatment prior to staining of paraffin sections. Sodium citrate buffer pH 6.0 is recommended for this purpose. . Western Blotting: Minimum Dilution: 2ug/ml; Maximum Dilution: 4ug/ml
other info1 :
Perservative Stabilisers: 0.02% Sodium Azide
other info2 :

ncbi summary :
This gene encodes a member of the BCL-2 family of proteins. This family member belongs to the BH3-only pro-apoptotic subclass. The protein cooperates with direct activator proteins to induce mitochondrial outer membrane permeabilization and apoptosis. It can bind to anti-apoptotic Bcl-2 family members to induce mitochondrial dysfunction and caspase activation. Because of its pro-apoptotic role, this gene is a potential drug target for cancer therapy and for tissue injury. Alternative splicing results in multiple transcript variants. uniprot summary :
PUMA: Essential mediator of p53/TP53-dependent and p53/TP53- independent apoptosis. Interacts with MCL1 and BCL2A1. Interacts with BCL2 and BCL2L1/BCL-XL. By DNA damage, glucocorticoid treatment, growth factor deprivation and p53/TP53. Ubiquitously expressed. Belongs to the Bcl-2 family. 4 isoforms of the human protein are produced by alternative splicing. Protein type: Mitochondrial. Chromosomal Location of Human Ortholog: 19q13.3-q13.4. Cellular Component: mitochondrial outer membrane; mitochondrion; cytosol. Molecular Function: protein binding. Biological Process: caspase activation; release of cytochrome c from mitochondria; apoptosis; positive regulation of protein homooligomerization; positive regulation of neuron apoptosis; release of sequestered calcium ion into cytosol; reduction of endoplasmic reticulum calcium ion concentration; determination of adult life span; response to DNA damage stimulus; negative regulation of growth
