specificity :
This item recognises an epitope of p53 between amino acids 156-214 which is present in some malignant tissues but not in normal and pre-malignant tissues. PAb240 is commonly reported as being mutant specific, however there are reports in the literature which suggest that conformational changes in the wild type may result in recognition by this clone.
form :
Purified. Purified IgG - liquid
concentration :
IgG concentration 0.1 mg/ml
storage stability :
Store at 4 degree C or at -20 degree C if preferred. This product should be stored undiluted. Storage in frost free freezers is not recommended. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use. Shelf Life: 18 months from date of despatch
tested application :
Immunoprecipitation (IP), Western Blot (WB)*
app notes :
Western Blot: Mutant p53 occurs because point mutations cause a conformational disruption. This exposes an internal epitope that This item is specific for. To be able to distinguish between wild and mutant forms of p53 non reduced conditions must be used. Western Blotting: Application Note: Non -reducing conditions are required for discrimination between wild and mutant forms of p53 using Pab240.
other info1 :
Perservative Stabilisers: 0.09% Sodium Azide (NaN3). 1% Bovine Serum Albumin. Preparation: Purified IgG prepared by affinity chromatography on Protein A from tissue culture supernatant
other info2 :
Immunogen: p53-beta-galactosidase fusion protein. Buffer Solution: Phosphate buffered saline. Target Species: Human

ncbi summary :
This gene encodes a tumor suppressor protein containing transcriptional activation, DNA binding, and oligomerization domains. The encoded protein responds to diverse cellular stresses to regulate expression of target genes, thereby inducing cell cycle arrest, apoptosis, senescence, DNA repair, or changes in metabolism. Mutations in this gene are associated with a variety of human cancers, including hereditary cancers such as Li-Fraumeni syndrome. Alternative splicing of this gene and the use of alternate promoters result in multiple transcript variants and isoforms. Additional isoforms have also been shown to result from the use of alternate translation initiation codons (PMIDs: 12032546, 20937277). [provided by RefSeq, Feb 2013]
