This item recognises Raf-1 phosphorylated at the serine residue at position 233. The antibody binds to Raf-1 that has been activated by cAMP activity at the newly identified serine 233 position, but not to resting, Raf-1. The antibody does not bind to Raf peptides phosphorylated at other sites.

Store at 4 degree C or at -20 degree C if preferred. This product should be stored undiluted. Storage in frost free freezers is not recommended. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use. Western Blotting: Minimum Dilution: 1/100; Maximum Dilution: 1/1000; Application Note: For the detection of phosphoproteins, serine phosphatase inhibitors such as 10mM Sodium Fluoride should be added to the sample buffer. Milk or other casein-based blocking solutions are not recommended as casein is a phosphoprotein and its use can result in high background.

This gene is the cellular homolog of viral raf gene (v-raf). The encoded protein is a MAP kinase kinase kinase (MAP3K), which functions downstream of the Ras family of membrane associated GTPases to which it binds directly. Once activated, the cellular RAF1 protein can phosphorylate to activate the dual specificity protein kinases MEK1 and MEK2, which in turn phosphorylate to activate the serine/threonine specific protein kinases, ERK1 and ERK2. Activated ERKs are pleiotropic effectors of cell physiology and play an important role in the control of gene expression involved in the cell division cycle, apoptosis, cell differentiation and cell migration.
