Approximate Protein Concentration: 1mg/ml
storage stability :
Store at 4 degree C or at -20 degree C if preferred. Storage in frost-free freezers is not recommended. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use. Shelf Life: 18 months from date of despatch.
tested application :
ELISA (EIA), Immunohistology Paraffin*, Western Blot (WB)
app notes :
ELISA: Minimum Dilution: 1/500; Maximum Dilution: 1/2000;. Immunohistology - Paraffin: Minimum Dilution: 1/100; Maximum Dilution: 1/400; Application Note: This antibody is suitable for use on paraffin embedded tissue sections. We do however recommend fixation in 95% ethanol/5% acetic acid for 2-3 hours prior to paraffin embedding. Specimens which have not been fixed in acetic acid/alcohol will require. pretreatment using the microwave-citrate buffer method.

products description :
Mouse anti Human Protein Gene Product 9.5 antibody, clone 31A3 recognizes protein gene product 9.5 (PGP9.5), a ubiquitin hydrolase which is widely expressed in neuronal tissues and represents 1-2% of total soluble brain proteins. PGP9.5, also known as ubiquitin C-terminal hydrolase 1 (UCHL-1), is involved in the regulation of the ubuiquitin pathway. Mouse anti Human Protein Gene Product 9.5 antibody, clone 31A3 stains neuronal cell bodies and axons in the CNS and periphery, small nerve fibres in peripheral tissues, neuroendocrine cells in the pituitary, thyroid, pancreas and tumours of the DNES. Clones 31A3 and 13C4 each recognise a different epitope towards the N-terminus of the protein. Mouse anti Human Protein Gene Product 9.5 antibody, clone 31A3 also recognizes PGP9.5 in other species, including rat and rabbit but evidence suggests it does not bind to PGP9.5 in guinea pigs (Wilson et al. 1988). Mouse anti Human Protein Gene Product 9.5 antibody, clone 31A3 has been used successfully as a capture reagent with clone 13C4 as a detection reagent in a sandwich ELISA to evaluate contamination of processed meat samples with neuronal tissue (Gauntiz et al. 2009).

ncbi summary :
The protein encoded by this gene belongs to the peptidase C12 family. This enzyme is a thiol protease that hydrolyzes a peptide bond at the C-terminal glycine of ubiquitin. This gene is specifically expressed in the neurons and in cells of the diffuse neuroendocrine system. Mutations in this gene may be associated with Parkinson disease.[provided by RefSeq, Sep 2009]
uniprot summary :
UCHL1: Ubiquitin-protein hydrolase involved both in the processing of ubiquitin precursors and of ubiquitinated proteins. This enzyme is a thiol protease that recognizes and hydrolyzes a peptide bond at the C-terminal glycine of ubiquitin. Also binds to free monoubiquitin and may prevent its degradation in lysosomes. The homodimer may have ATP-independent ubiquitin ligase activity. Monomer. Homodimer. Interacts with SNCA. Interacts with COPS5. Found in neuronal cell bodies and processes throughout the neocortex. Expressed in neurons and cells of the diffuse neuroendocrine system and their tumors. Weakly expressed in ovary. Down-regulated in brains from Parkinson disease and Alzheimer disease patients. Belongs to the peptidase C12 family. Protein type: Ligase; EC 3.4.19.12; Cell development/differentiation; Ubiquitin conjugating system; Protease. Chromosomal Location of Human Ortholog: 4p14. Cellular Component: nucleoplasm; endoplasmic reticulum membrane; cell soma; axon; cytoplasm; plasma membrane; cytosol. Molecular Function: protein binding; ubiquitin binding; omega peptidase activity; cysteine-type endopeptidase activity; ubiquitin-specific protease activity; alpha-2A adrenergic receptor binding; ligase activity. Biological Process: cell proliferation; negative regulation of MAP kinase activity; proteasomal ubiquitin-dependent protein catabolic process; axon transport of mitochondrion; protein deubiquitination; axon target recognition; eating behavior; neuromuscular process; sensory perception of pain; muscle fiber development; adult walking behavior.
