Expression System: E Coli. Bioactivity: Specific activity is > 20,000pmol/min/ug, and was obtained by measuring the decrease of NAD in absorbance at 340nm resulting from NADH at pH 6.5 at 37C. Activity Assay: 1. Prepare 180ul assay buffer into a suitable container: The concentrations are 80 mM Triethanolamine (pH 6.5), 0.3 mM 2-phosphorglycerate, 0.2 mM beta- NADH, 25 mM magnesium sulfate, 100 mM potassium chloride, 1.2 mM ADP, 3.5 unit pyruvate kinase, 5 unit L-lactic dehydrogenas. 2. Equilibrate to 37C and monitor at A340nm until the value is constant using a spectrophotometer. 3. Add 20ul of recombinant a-enolase protein 10ug/ml in assay buffer. 4. Record the decrease in A340nm for 5 minutes at 37C.

products description :
Alpha-enolase, also known as Enolase 1, is one of three enolase isoenzymes and a glycolytic enzyme expressed in most tissues. This protein plays a key role in anaerobic metabolism under hypoxic conditions and may act as a cell surface plasminogen receptor during tissue invasion. Abnormal expression of alpha-enolase is associated with tumor progression in some cases of breast and lung cancer. It also has been identified as an autoantigen associated with Hashimoto's encephalopathy and severe asthma. Recombinant human alpha-enolase was expressed in E.coli and purified by using conventional chromatography.

ncbi summary :
This gene encodes alpha-enolase, one of three enolase isoenzymes found in mammals. Each isoenzyme is a homodimer composed of 2 alpha, 2 gamma, or 2 beta subunits, and functions as a glycolytic enzyme. Alpha-enolase in addition, functions as a structural lens protein (tau-crystallin) in the monomeric form. Alternative splicing of this gene results in a shorter isoform that has been shown to bind to the c-myc promoter and function as a tumor suppressor. Several pseudogenes have been identified, including one on the long arm of chromosome 1. Alpha-enolase has also been identified as an autoantigen in Hashimoto encephalopathy.
