NAD-dependent deacetylase sirtuin-3, mitochondrial, also known as SIRT3, belongs to the sirtuin family of proteins. Members of the sirtuin family are characterized by a sirtuin core domain and grouped into four classes. The functions of human sirtuins have a range of molecular functions and have emerged as important proteins in aging, stress resistance and metabolic regulation. SIRT3 exhibits NAD+-dependent deacetylase activity in the mitochondria. Over-expression of SIRT3 results in increased levels of the mitochondrial uncoupling protein 1. SIRT3 protein levels are also elevated in certain breast cancers. Recombinant human SIRT3 protein, fused to His-tag at N-terminus, was expressed in E.coli and purified by using conventional chromatography techniques.

This gene encodes a member of the sirtuin family of proteins, homologs to the yeast Sir2 protein. Members of the sirtuin family are characterized by a sirtuin core domain and grouped into four classes. The functions of human sirtuins have not yet been determined; however, yeast sirtuin proteins are known to regulate epigenetic gene silencing and suppress recombination of rDNA. Studies suggest that the human sirtuins may function as intracellular regulatory proteins with mono-ADP-ribosyltransferase activity. The protein encoded by this gene is included in class I of the sirtuin family. Two alternatively spliced transcript variants that encode different proteins have been described for this gene.
