Expression System: E Coli. Bioactivity: Specific activity: approximately 0.25-0.3 units/mg. Enzymatic activity was confirmed by measuring the amount of enzyme catalyzing the oxidation of 1 micromole NADPH per minute at 25C. Specific activity was expressed as units/mg protein. Assay: 1. Prepare a 1ml reaction mix into a suitable container: The final concentrations are 0.1M sodium phosphate (pH7.0), 10mM DL-glyceraldehyde, 0.3mM NADPH. 2. Add 50ul of recombinant AKR7A2 solution with various concentrations (2.5ug, 5ug, 10ug) in 750ul reaction buffer. 3. Mix by inversion and Incubate at 25C for 2.5 minutes. 4. Add 200ul of 50 mM DL-glyceraldehyde as a substrate and immediately mix by inversion. 5. Record the increase at A340nm for 3 minutes.

products description :
Aldo-keto reductases, such as AKR7A2, are involved in the detoxification of aldehydes and ketones. This protein can reduce the dialdehyde protein-binding form of aflatoxin B1 (AFB1) to the nonbinding AFB1 dialcohol. It may be involved in protection of liver against the toxic and carcinogenic effects of AFB1, a potent hepatocarcinogen. Also, it has been proposed previously to catalyze the NADPH-dependent reduction of succinic semialdehyde (SSA) to gamma-Hydroxybutyrate in human brain. Recombinant AKR7A2 protein was expressed in E.coli and purified by using conventional chromatography techniques.

ncbi summary :
The protein encoded by this gene belongs to the aldo/keto reductase (AKR) superfamily and AKR7 family, which are involved in the detoxification of aldehydes and ketones. The AKR7 family consists of 3 genes that are present in a cluster on the p arm of chromosome 1. This protein, thought to be localized in the golgi, catalyzes the NADPH-dependent reduction of succinic semialdehyde to the endogenous neuromodulator, gamma-hydroxybutyrate. It may also function as a detoxication enzyme in the reduction of aflatoxin B1 and 2-carboxybenzaldehyde. [provided by RefSeq, Oct 2011]
uniprot summary :
AKR7A2: Catalyzes the NADPH-dependent reduction of succinic semialdehyde to gamma-hydroxybutyrate. May have an important role in producing the neuromodulator gamma-hydroxybutyrate (GHB). Has broad substrate specificity. Has NADPH-dependent aldehyde reductase activity towards 2-carboxybenzaldehyde, 2- nitrobenzaldehyde and pyridine-2-aldehyde (in vitro). Can reduce 1,2-naphthoquinone and 9,10-phenanthrenequinone (in vitro). Can reduce the dialdehyde protein-binding form of aflatoxin B1 (AFB1) to the non-binding AFB1 dialcohol. May be involved in protection of liver against the toxic and carcinogenic effects of AFB1, a potent hepatocarcinogen. Belongs to the aldo/keto reductase family. Aldo/keto reductase 2 subfamily. Protein type: EC 1.1.1.n11; Oxidoreductase. Chromosomal Location of Human Ortholog: 1p36.13. Cellular Component: Golgi apparatus; mitochondrion; cytosol. Molecular Function: electron carrier activity; aldehyde reductase activity; phenanthrene-9,10-epoxide hydrolase activity. Biological Process: xenobiotic metabolic process; carbohydrate metabolic process; aldehyde metabolic process
