SDF-1 is small cytokine belonging to the chemokine family that is officially designated Chemokine (C-X-C motif) ligand 12 (CXCL12). SDF-1 is strongly chemotactic for lymphocytes and works by its receptor CXCR4. The SDF-1+CXCR4 complex plays a significant role in the creation of metastases of neoplasms and as a response to cytostatic treatment. Identification of this complex may be a useful prognostic factor in the therapy of many types of carcinoma. Recombinant human SDF-1, fused to His-tag at N-terminus, was expressed in E.coli and purified by using conventional chromatography.

Mutations in this gene are associated with resistance to human immunodeficiency virus type 1 infections. Multiple transcript variants encoding different isoforms have been found for this gene. Activates the C-X-C chemokine receptor CXCR4 to induce a rapid and transient rise in the level of intracellular calcium ions and chemotaxis. Also binds to another C-X-C chemokine receptor CXCR7, which activates the beta-arrestin pathway and acts as a scavenger receptor for SDF-1. SDF-1-beta(3-72) and SDF-1- alpha(3-67) show a reduced chemotactic activity.
