uniprot entry name :
TNF11_HUMAN
specificity :
This assay has high sensitivity and excellent specificity for detection of RANkL. No significant cross-reactivity or interference between RANkL and analogues was observed.
storage stability :
The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.

products description :
Intended Uses: The kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of RANkL in gallus serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. Principle of the Assay: The microplate provided in this kit has been pre-coated with an antibody specific to RANkL. Standards or samples are then added to the appropriate microplate wells with a biotin-conjugated antibody specific to RANkL. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain RANkL, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of RANkL in the samples is then determined by comparing the O.D. of the samples to the standard curve.

ncbi summary :
This gene encodes a member of the tumor necrosis factor (TNF) cytokine family which is a ligand for osteoprotegerin and functions as a key factor for osteoclast differentiation and activation. This protein was shown to be a dentritic cell survival factor and is involved in the regulation of T cell-dependent immune response. T cell activation was reported to induce expression of this gene and lead to an increase of osteoclastogenesis and bone loss. This protein was shown to activate antiapoptotic kinase AKT/PKB through a signaling complex involving SRC kinase and tumor necrosis factor receptor-associated factor (TRAF) 6, which indicated this protein may have a role in the regulation of cell apoptosis. Targeted disruption of the related gene in mice led to severe osteopetrosis and a lack of osteoclasts. The deficient mice exhibited defects in early differentiation of T and B lymphocytes, and failed to form lobulo-alveolar mammary structures during pregnancy. Two alternatively spliced transcript variants have been found. [provided by RefSeq, Jul 2008]
uniprot summary :
TNFSF11: Cytokine that binds to TNFRSF11B/OPG and to TNFRSF11A/RANK. Osteoclast differentiation and activation factor. Augments the ability of dendritic cells to stimulate naive T-cell proliferation. May be an important regulator of interactions between T-cells and dendritic cells and may play a role in the regulation of the T-cell-dependent immune response. May also play an important role in enhanced bone-resorption in humoral hypercalcemia of malignancy. Homotrimer. Up-regulated by T-cell receptor stimulation. Highest in the peripheral lymph nodes, weak in spleen, peripheral blood Leukocytes, bone marrow, heart, placenta, skeletal muscle, stomach and thyroid. Belongs to the tumor necrosis factor family. 3 isoforms of the human protein are produced by alternative splicing. Protein type: Membrane protein, integral. Chromosomal Location of Human Ortholog: 13q14. Cellular Component: extracellular space; integral to plasma membrane; cytoplasm; extracellular region. Molecular Function: cytokine activity; tumor necrosis factor receptor superfamily binding; tumor necrosis factor receptor binding.
