specificity :
This assay has high sensitivity and excellent specificity for detection of Kallikrein 14 (KLK14). No significant cross-reactivity or interference between Kallikrein 14 (KLK14) and analogues was observed.
storage stability :
For unopened kit: All the reagents should be kept according to the labels on vials. The Standard, Detection Reagent A, Detection Reagent B and the 96-well strip plate should be stored at -20 degree C upon receipt while the others should be at 4 degree C. For opened kit: When the kit is opened, the remaining reagents still need to be stored according to the above storage condition. Besides, please return the unused wells to the foil pouch containing the desiccant pack, and reseal along entire edge of zip-seal. The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.
other info1 :
Samples: Serum, plasma, tissue homogenates and other biological fluids. Assay Type: Competitive. Detection Range: 123.46-10000pg/mL. Sensitivity: Typically less than 43.2pg/mL.
other info2 :
Precision: Intra-assay Precision (Precision within an assay): 3 samples with low, middle and high level Kallikrein 14 (KLK14) were tested 20 times on one plate, respectively. Inter-assay Precision (Precision between assays): 3 samples with low, middle and high level Kallikrein 14 (KLK14) were tested on 3 different plates, 8 replicates in each plate. CV(%) = SD/meanX100 . Intra-Assay: CV<10% . Inter-Assay: CV<12% . Assay Procedure Summary: 1. Prepare all reagents, samples and standards;. 2. Add 50uL standard or sample to each well. And then add 50uL prepared Detection Reagent A immediately. Shake and mix. Incubate 1 hour at 37 degree C;. 3. Aspirate and wash 3 times;. 4. Add 100uL prepared Detection Reagent B. Incubate 30 minutes at 37 degree C;. 5. Aspirate and wash 5 times;. 6. Add 90uL Substrate Solution. Incubate 15-25 minutes at 37 degree C;. 7. Add 50uL Stop Solution. Read at 450 nm immediately.
products description :
Principle of the Assay: This assay employs the competitive inhibition enzyme immunoassay technique. A monoclonal antibody specific to Kallikrein 14 (KLK14) has been pre-coated onto a microplate. A competitive inhibition reaction is launched between biotin labeled Kallikrein 14 (KLK14) and unlabeled Kallikrein 14 (KLK14) (Standards or samples) with the pre-coated antibody specific to Kallikrein 14 (KLK14). After incubation the unbound conjugate is washed off. Next, avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. The amount of bound HRP conjugate is reverse proportional to the concentration of Kallikrein 14 (KLK14) in the sample. After addition of the substrate solution, the intensity of color developed is reverse proportional to the concentration of Kallikrein 14 (KLK14) in the sample.

ncbi summary :
This gene encodes a member of the kallikrein subfamily of serine proteases that have diverse physiological functions such as regulation of blood pressure and desquamation. The altered expression of this gene is implicated in the progression of different cancers including breast and prostate tumors. The encoded protein is a precursor that is proteolytically processed to generate the functional enzyme. This gene is one of the fifteen kallikrein subfamily members located in a cluster on chromosome 19. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Jul 2015]
uniprot summary :
KLK14: Serine-type endopeptidase with a dual trypsin-like and chymotrypsin-like substrate specificity. May activate/inactivate the proteinase-activated receptors F2R, F2RL1 and F2RL3 and other kallikreins including KLK1, KLK3, KLK5 and KLK11. May function in seminal clot liquefaction through direct cleavage of the semenogelin SEMG1 and SEMG2 and activation of KLK3. May function through desmoglein DSG1 cleavage in epidermal desquamation a process by which the most superficial corneocytes are shed from the skin surface. May be involved in several aspects of tumor progression including growth, invasion and angiogenesis. Belongs to the peptidase S1 family. Kallikrein subfamily. Protein type: Secreted; EC 3.4.21.-; Secreted, signal peptide; Protease. Chromosomal Location of Human Ortholog: 19q13.3-q13.4. Cellular Component: extracellular space. Molecular Function: serine-type endopeptidase activity. Biological Process: fertilization; negative regulation of G-protein coupled receptor protein signaling pathway; positive regulation of G-protein coupled receptor protein signaling pathway; epidermis morphogenesis; proteolysis
