specificity :
This assay has high sensitivity and excellent specificity for detection of TDGF1. No significant cross-reactivity or interference between TDGF1 and analogues was observed.
storage stability :
For unopened kit: All the reagents should be kept according to the labels on vials. The Standard, Detection Reagent A, Detection Reagent B and the 96-well strip plate should be stored at -20 degree C upon receipt while the others should be at 4 degree C. For opened kit: When the kit is opened, the remaining reagents still need to be stored according to the above storage condition. Besides, please return the unused wells to the foil pouch containing the desiccant pack, and reseal along entire edge of zip-seal. The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.
other info1 :
Samples: Human serum, plasma, tissue homogenates, cell lysates and other biological fluids. Assay Type: Sandwich. Detection Range: 78-5,000pg/mL. The standard curve concentrations used for the ELISA's were 5,000pg/mL, 2,500pg/mL, 1,250pg/mL, 625pg/mL, 312pg/mL, 156pg/mL, 78pg/mL. Sensitivity: 28pg/mL.
other info2 :
Intra-assay Precision: Intra-assay Precision (Precision within an assay): 3 samples with low, middle and high level TDGF1 were tested 20 times on one plate, respectively. Intra-Assay: CV<10%. Inter-assay Precision: Inter-assay Precision (Precision between assays): 3 samples with low, middle and high level TDGF1 were tested on 3 different plates, 8 replicates in each plate. CV(%) = SD/meanX100. Inter-Assay: CV<12%
products description :
Intended Uses: The kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of TDGF1 in human serum, plasma, tissue homogenates, cell lysates and other biological fluids. Principle of the Assay The microtiter plate provided in this kit has been pre-coated with an antibody specific to TDGF1. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to TDGF1. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain TDGF1, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of TDGF1 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

ncbi summary :
This gene encodes an epidermal growth factor-related protein that contains a cripto, FRL-1, and cryptic domain. The encoded protein is an extracellular, membrane-bound signaling protein that plays an essential role in embryonic development and tumor growth. Mutations in this gene are associated with forebrain defects. Pseudogenes of this gene are found on chromosomes 2, 3, 6, 8, 19 and X. Alternate splicing results in multiple transcript variants. uniprot summary :
TDGF1: Could play a role in the determination of the epiblastic cells that subsequently give rise to the mesoderm. Interacts with the activin type-1 receptor ACVR1B. Preferentially expressed in gastric and colorectal carcinomas than in their normal counterparts. Expressed in breast and lung. Protein type: Membrane protein, GPI anchor; Membrane protein, peripheral; Motility/polarity/chemotaxis; Cell adhesion; Cell cycle regulation. Chromosomal Location of Human Ortholog: 3p21.31. Cellular Component: extrinsic to plasma membrane; Golgi apparatus; extracellular space; cell surface; perinuclear region of cytoplasm; apical plasma membrane; plasma membrane; nucleus; lipid raft.
