specificity :
This assay has high sensitivity and excellent specificity for detection of GDNF. No significant cross-reactivity or interference between GDNF and analogues was observed.
storage stability :
The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.

products description :
Intended Uses: The kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of GDNF in mouse serum, plasma, tissue homogenates, cell lysates, cerebrospinal fluid, cell culture supernates and other biological fluids. Principle of the Assay: The microplate provided in this kit has been pre-coated with an antibody specific to GDNF. Standards or samples are then added to the appropriate microplate wells with a biotin-conjugated antibody specific to GDNF. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain GDNF, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of GDNF in the samples is then determined by comparing the O.D. of the samples to the standard curve.

uniprot summary :
Gelsolin: a calcium-regulated, actin-modulating protein that binds to the plus (or barbed) ends of actin monomers or filaments, preventing monomer exchange (end-blocking or capping). It can promote the assembly of monomers into filaments (nucleation) as well as sever filaments already formed. Its expression is significantly decreased in many tumors and transformed cell lines. Gelsolin and other proteins including E-cadherin, BRCA, p21WAF, HoxA5, and retinoic acid receptor II are regulated by epigenetic changes in breast cancers. Gelsolin helps mediate the ability of HDAC inhibitors to protect neurons from oxygen/glucose deprivation. Two human isoforms are produced by alternative initiation. Protein type: Nuclear receptor co-regulator; Motility/polarity/chemotaxis; Actin-binding; Secreted, signal peptide; Secreted. Cellular Component: ruffle; extracellular space; protein complex; focal adhesion; perinuclear region of cytoplasm; lamellipodium; extracellular region; cytosol; actin cytoskeleton. Molecular Function: calcium ion binding; actin binding. Biological Process: regulation of cell adhesion; actin filament severing; phosphoinositide-mediated signaling; wound healing; apoptosis; actin nucleation; vesicle-mediated transport; response to ethanol; tissue regeneration; actin filament polymerization; response to folic acid; actin polymerization and/or depolymerization; barbed-end actin filament capping; oligodendrocyte development; aging
