specificity :
This assay has high sensitivity and excellent specificity for detection of MAPRE1. No significant cross-reactivity or interference between MAPRE1 and analogues was observed.
storage stability :
For unopened kit: All the reagents should be kept according to the labels on vials. The Standard, Detection Reagent A, Detection Reagent B and the 96-well strip plate should be stored at -20 degree C upon receipt while the others should be at 4 degree C. For opened kit: When the kit is opened, the remaining reagents still need to be stored according to the above storage condition. Besides, please return the unused wells to the foil pouch containing the desiccant pack, and reseal along entire edge of zip-seal. The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.
other info1 :
Samples: Human tissue homogenates, cell lysates and other biological fluids. Assay Type: Sandwich. Detection Range: 0.156-10ng/mL. Sensitivity: 0.058ng/mL
other info2 :
Intra-assay Precision: Intra-assay Precision (Precision within an assay): 3 samples with low, middle and high level MAPRE1 were tested 20 times on one plate, respectively. Intra-Assay: CV<10%. Inter-assay Precision: Inter-assay Precision (Precision between assays): 3 samples with low, middle and high level MAPRE1 were tested on 3 different plates, 8 replicates in each plate. CV(%) = SD/meanX100. Inter-Assay: CV<12%
products description :
Intended Uses: The kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of MAPRE1 in human tissue homogenates, cell lysates and other biological fluids. Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to MAPRE1. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to MAPRE1. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain MAPRE1, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of MAPRE1 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

ncbi summary :
The protein encoded by this gene was first identified by its binding to the APC protein which is often mutated in familial and sporadic forms of colorectal cancer. This protein localizes to microtubules, especially the growing ends, in interphase cells. During mitosis, the protein is associated with the centrosomes and spindle microtubules. The protein also associates with components of the dynactin complex and the intermediate chain of cytoplasmic dynein. Because of these associations, it is thought that this protein is involved in the regulation of microtubule structures and chromosome stability. This gene is a member of the RP/EB family. uniprot summary :
EB1: a microtubule-associated protein of the RP/EB family. Binds to the APC protein which is often mutated in familial and sporadic forms of colorectal cancer. Localizes to microtubules, especially the growing ends, in interphase cells. Associates with the centrosomes and spindle microtubules during mitosis. Also associates with components of the dynactin complex and the intermediate chain of cytoplasmic dynein. May be involved in the regulation of microtubule structures and chromosome stability. Protein type: Motility/polarity/chemotaxis; Cytoskeletal. Chromosomal Location of Human Ortholog: 20q11.1-q11.23. Cellular Component: Golgi apparatus; microtubule; centrosome; cytoplasmic microtubule; cell projection membrane; cytoplasm; cortical microtubule cytoskeleton; spindle; cytosol. Molecular Function: protein C-terminus binding; microtubule plus-end binding; protein binding. Biological Process: mitosis; cell proliferation; cell division; organelle organization and biogenesis; negative regulation of microtubule polymerization; mitotic cell cycle; G2/M transition of mitotic cell cycle
