specificity :
This assay has high sensitivity and excellent specificity for detection of STIP1. No significant cross-reactivity or interference between STIP1 and analogues was observed.
storage stability :
For unopened kit: All the reagents should be kept according to the labels on vials. The Standard, Detection Reagent A, Detection Reagent B and the 96-well strip plate should be stored at -20 degree C upon receipt while the others should be at 4 degree C. For opened kit: When the kit is opened, the remaining reagents still need to be stored according to the above storage condition. Besides, please return the unused wells to the foil pouch containing the desiccant pack, and reseal along entire edge of zip-seal. The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.
other info1 :
Samples: Human serum, plasma, tissue homogenates and other biological fluids. Assay Type: Sandwich. Detection Range: 1.56-100ng/mL. The standard curve concentrations used for the ELISA's were 100ng/mL, 50ng/mL, 25ng/mL, 12.5ng/mL, 6.25ng/mL, 3.12ng/mL, 1.56ng/mL. Sensitivity: 0.63ng/mL.
other info2 :
Intra-assay Precision: Intra-assay Precision (Precision within an assay): 3 samples with low, middle and high level STIP1 were tested 20 times on one plate, respectively. Intra-Assay: CV<10%. Inter-assay Precision: Inter-assay Precision (Precision between assays): 3 samples with low, middle and high level STIP1 were tested on 3 different plates, 8 replicates in each plate. CV(%) = SD/meanX100. Inter-Assay: CV<12%
products description :
Intended Uses: The kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of STIP1 in human serum, plasma, tissue homogenates and other biological fluids. Principle of the Assay The microtiter plate provided in this kit has been pre-coated with an antibody specific to STIP1. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to STIP1. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain STIP1, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of STIP1 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

ncbi summary :
STIP1 is an adaptor protein that coordinates the functions of HSP70 (see HSPA1A; MIM 140550) and HSP90 (see HSP90AA1; MIM 140571) in protein folding. It is thought to assist in the transfer of proteins from HSP70 to HSP90 by binding both HSP90 and substrate-bound HSP70. STIP1 also stimulates the ATPase activity of HSP70 and inhibits the ATPase activity of HSP90, suggesting that it regulates both the conformations and ATPase cycles of these chaperones (Song and Masison, 2005 [PubMed 16100115]).[supplied by OMIM, Jul 2009]
uniprot summary :
STI1: a protein that is apparently involved in the response to stress. Specifically interacts with the C-terminal tails of Hsp70 and Hsp90 via TPR1 and TPR2A domains. Deletion of STI1 reduces the activity of the glucocorticoid receptor, a Hsp90 target protein. Phosphorylated in vitro by p90RSK and casein kinase II. Protein type: Chaperone. Chromosomal Location of Human Ortholog: 11q13. Cellular Component: Golgi apparatus; nucleus. Molecular Function: protein binding. Biological Process: response to stress
