This assay has high sensitivity for detection of UCHL1. 100% cross-reactivity of UCHL1 was observed among human, mouse and rat.
storage stability :
For unopened kit, all the reagents should be kept according to the label on each vial. The Standard, Detection Reagent A, Detection Reagent B and the 96-well strip plate should be stored at -20 degree C while others should be at 4 degree C.
other info1 :
Samples: Tissue homogenates, cell lysates, cell culture supernates and other biological fluids. Due to the 96% homology of the sequence among different species, the kit can be used to detect human, mouse and rat samples. Assay Type: Sandwich. Detection Range: 0.78 -50ng/mL. The standard curve concentrations used for the ELISA's were 50ng/mL, 25ng/mL, 12.5ng/mL, 6.25ng/mL, 3.12ng/mL, 1.56ng/mL, 0.78ng/mL. Sensitivity: 0.28ng/mL.
other info2 :
Intra-assay Precision: Intra-assay Precision (Precision within an assay): 3 samples with low, middle and high level UCHL1 were tested 20 times on one plate, respectively. Intra-Assay: CV<10%. Inter-assay Precision: Inter-assay Precision (Precision between assays): 3 samples with low, middle and high level UCHL1 were tested on 3 different plates, 8 replicates in each plate. CV(%) = SD/meanX100. Inter-Assay: CV<12%
products description :
Intended Uses: The kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of UCHL1 in tissue homogenates, cell lysates, cell culture supernates and other biological fluids. Due to the 96% homology of the sequence among different species, the kit can be used to detect human, mouse and rat samples. Principle of the Assay The microtiter plate provided in this kit has been pre-coated with an antibody specific to UCHL1. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to UCHL1. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain UCHL1, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of UCHL1 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

ncbi summary :
The protein encoded by this gene belongs to the peptidase C12 family. This enzyme is a thiol protease that hydrolyzes a peptide bond at the C-terminal glycine of ubiquitin. This gene is specifically expressed in the neurons and in cells of the diffuse neuroendocrine system. Mutations in this gene may be associated with Parkinson disease.[provided by RefSeq, Sep 2009]
uniprot summary :
UCHL1: Ubiquitin-protein hydrolase involved both in the processing of ubiquitin precursors and of ubiquitinated proteins. This enzyme is a thiol protease that recognizes and hydrolyzes a peptide bond at the C-terminal glycine of ubiquitin. Also binds to free monoubiquitin and may prevent its degradation in lysosomes. The homodimer may have ATP-independent ubiquitin ligase activity. Monomer. Homodimer. Interacts with SNCA. Interacts with COPS5. Found in neuronal cell bodies and processes throughout the neocortex. Expressed in neurons and cells of the diffuse neuroendocrine system and their tumors. Weakly expressed in ovary. Down-regulated in brains from Parkinson disease and Alzheimer disease patients. Belongs to the peptidase C12 family. Protein type: Protease; Cell development/differentiation; EC 3.4.19.12; Ligase; Ubiquitin conjugating system. Chromosomal Location of Human Ortholog: 4p14. Cellular Component: nucleoplasm; endoplasmic reticulum membrane; cell soma; axon; cytoplasm; plasma membrane; cytosol. Molecular Function: protein binding; ubiquitin binding; omega peptidase activity; cysteine-type endopeptidase activity; ubiquitin-specific protease activity; alpha-2A adrenergic receptor binding; ligase activity. Biological Process: negative regulation of MAP kinase activity; cell proliferation; proteasomal ubiquitin-dependent protein catabolic process; axon target recognition; axon transport of mitochondrion; protein deubiquitination; eating behavior; neuromuscular process; muscle fiber development; sensory perception of pain; adult walking behavior. Disease: Neurodegeneration With Optic Atrophy, Childhood-onset; Parkinson Disease 5, Autosomal Dominant
