specificity :
This assay has high sensitivity and excellent specificity for detection of Chemokine C-X3-C-Motif Receptor 1 (CX3CR1). No significant cross-reactivity or interference between Chemokine C-X3-C-Motif Receptor 1 (CX3CR1) and analogues was observed.
storage stability :
For unopened kit: All the reagents should be kept according to the labels on vials. The Standard, Detection Reagent A, Detection Reagent B and the 96-well strip plate should be stored at -20 degree C upon receipt while the others should be at 4 degree C. For opened kit: When the kit is opened, the remaining reagents still need to be stored according to the above storage condition. Besides, please return the unused wells to the foil pouch containing the desiccant pack, and reseal along entire edge of zip-seal. The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.
other info1 :
Samples: Tissue homogenates, Cell lysates and other biological fluids. Assay Type: Sandwich. Detection Range: 0.156-10ng/mL. Sensitivity: Typically less than 0.054ng/mL.

Incubate 2 hours at 37 degree C;. 3. Aspirate and add 100uL prepared Detection Reagent A. Incubate 1 hour at 37 degree C;. 4. Aspirate and wash 3 times;. 5. Add 100uL prepared Detection Reagent B. Incubate 30 minutes at 37 degree C;. 6. Aspirate and wash 5 times;. 7. Add 90uL Substrate Solution. Incubate 15-25 minutes at 37 degree C;. 8. Add 50uL Stop Solution. Read at 450nm immediately.
products description :
Principle of the Assay: The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Chemokine C-X3-C-Motif Receptor 1 (CX3CR1). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Chemokine C-X3-C-Motif Receptor 1 (CX3CR1). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Chemokine C-X3-C-Motif Receptor 1 (CX3CR1), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm 10nm. The concentration of Chemokine C-X3-C-Motif Receptor 1 (CX3CR1) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

ncbi summary :
Fractalkine is a transmembrane protein and chemokine involved in the adhesion and migration of leukocytes. The protein encoded by this gene is a receptor for fractalkine. The encoded protein also is a coreceptor for HIV-1, and some variations in this gene lead to increased susceptibility to HIV-1 infection and rapid progression to AIDS. Four transcript variants encoding two different isoforms have been found for this gene. [provided by RefSeq, Jan 2010]
uniprot summary :
CX3CR1: Receptor for the CX3C chemokine fractalkine and mediates both its adhesive and migratory functions. Acts as coreceptor with CD4 for HIV-1 virus envelope protein (in vitro). Isoform 2 and isoform 3 seem to be more potent HIV-1 coreceptors than isoform 1. Defects in CX3CR1 are a cause of susceptibility to age- related macular degeneration type 12 (ARMD12). ARMD12 is a form of age-related macular degeneration, a multifactorial eye disease and the most common cause of irreversible vision loss in the developed world. In most patients, the disease is manifest as ophthalmoscopically visible yellowish accumulations of protein and lipid that lie beneath the retinal pigment epithelium and within an elastin-containing structure known as Bruch membrane. Belongs to the G-protein coupled receptor 1 family. 3 isoforms of the human protein are produced by alternative splicing. Protein type: Membrane protein, multi-pass; Membrane protein, integral; GPCR, family 1; Receptor, GPCR; Motility/polarity/chemotaxis; Cell adhesion; Receptor, cytokine. Chromosomal Location of Human Ortholog: 3p21.3. Cellular Component: neuron projection; integral to plasma membrane; perinuclear region of cytoplasm; plasma membrane. Molecular Function: protein binding; chemokine receptor activity; C-X3-C chemokine receptor activity.
