specificity :
This assay has high sensitivity and excellent specificity for detection of synaptophysin. No significant cross-reactivity or interference between synaptophysin and analogues was observed.
storage stability :
For unopened kit: All the reagents should be kept according to the labels on vials. The Standard, Detection Reagent A, Detection Reagent B and the 96-well strip plate should be stored at -20 degree C upon receipt while the others should be at 4 degree C. For opened kit: When the kit is opened, the remaining reagents still need to be stored according to the above storage condition. Besides, please return the unused wells to the foil pouch containing the desiccant pack, and reseal along entire edge of zip-seal. The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.
other info1 :
Samples: Mouse serum, plasma, tissue homogenates, cerebrospinal fluid and other biological fluids. Assay Type: Sandwich. Detection Range: 0.625-40ng/mL. The standard curve concentrations used for the ELISA's were 40ng/mL, 20ng/mL, 10ng/mL, 5ng/mL, 2.5ng/mL, 1.25ng/mL, 0.625ng/mL. Sensitivity: 0.276ng/mL.
other info2 :
Intra-assay Precision: Intra-assay Precision (Precision within an assay): 3 samples with low, middle and high level synaptophysin were tested 20 times on one plate, respectively. Intra-Assay: CV<10%. Inter-assay Precision: Inter-assay Precision (Precision between assays): 3 samples with low, middle and high level synaptophysin were tested on 3 different plates, 8 replicates in each plate. CV(%) = SD/meanX100. Inter-Assay: CV<12%
products description :
Intended Uses: The kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of synaptophysin in mouse serum, plasma, tissue homogenates, cerebrospinal fluid and other biological fluids. Principle of the Assay The microtiter plate provided in this kit has been pre-coated with an antibody specific to synaptophysin. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to synaptophysin. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain synaptophysin, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of synaptophysin in the samples is then determined by comparing the O.D. of the samples to the standard curve.

ncbi summary :
This gene encodes an integral membrane protein of small synaptic vesicles in brain and endocrine cells. The protein also binds cholesterol and is thought to direct targeting of vesicle-associated membrane protein 2 (synaptobrevin) to intracellular compartments. Mutations in this gene are associated with X-linked mental retardation (XLMR). [provided by RefSeq, Aug 2011]
uniprot summary :
SYP: Possibly involved in structural functions as organizing other membrane components or in targeting the vesicles to the plasma membrane. Involved in the regulation of short-term and long-term synaptic plasticity. Homohexamer or homotetramer. Interacts with SRCIN1. Characteristic of a type of small (30-80 nm) neurosecretory vesicles, including presynaptic vesicles, but also vesicles of various neuroendocrine cells of both neuronal and epithelial phenotype. Belongs to the synaptophysin/synaptobrevin family. Protein type: Membrane protein, multi-pass; Membrane protein, integral; Vesicle. Chromosomal Location of Human Ortholog: Xp11.23-p11.22. Cellular Component: presynaptic membrane; synaptic vesicle; neuron projection; protein complex; synaptic vesicle membrane; presynaptic active zone; excitatory synapse; cell junction; integral to synaptic vesicle membrane. Molecular Function: identical protein binding; protein self-association; transporter activity; cholesterol binding; protein complex binding; syntaxin-1 binding; SH2 domain binding. Biological Process: regulation of long-term neuronal synaptic plasticity; synaptic vesicle maturation; synaptic vesicle membrane organization and biogenesis; endocytosis; regulation of short-term neuronal synaptic plasticity; regulation of neuronal synaptic plasticity. Disease: Mental Retardation, X-linked 96
