specificity :
This assay has high sensitivity and excellent specificity for detection of IL1RA. No significant cross-reactivity or interference between IL1RA and analogues was observed.
storage stability :
For unopened kit: All the reagents should be kept according to the labels on vials. The Standard, Detection Reagent A, Detection Reagent B and the 96-well strip plate should be stored at -20 degree C upon receipt while the others should be at 4 degree C. For opened kit: When the kit is opened, the remaining reagents still need to be stored according to the above storage condition. Besides, please return the unused wells to the foil pouch containing the desiccant pack, and reseal along entire edge of zip-seal. The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.
other info1 :
Samples: Mouse serum, plasma, tissue homogenates and other biological fluids. Assay Type: Sandwich. Detection Range: 3.12 -200pg/mL. The standard curve concentrations used for the ELISA's were 200pg/mL, 100pg/mL, 50pg/mL, 25pg/mL, 12.5pg/mL, 6.25pg/mL, 3.12pg/mL. Sensitivity: 1.29pg/mL.
other info2 :
Intra-assay Precision: Intra-assay Precision (Precision within an assay): 3 samples with low, middle and high level IL1RA were tested 20 times on one plate, respectively. Intra-Assay: CV<10%. Inter-assay Precision: Inter-assay Precision (Precision between assays): 3 samples with low, middle and high level IL1RA were tested on 3 different plates, 8 replicates in each plate. CV(%) = SD/meanX100. Inter-Assay: CV<12%
products description :
Intended Uses: The kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of IL1RA in mouse serum, plasma, tissue homogenates and other biological fluids. Principle of the Assay The microtiter plate provided in this kit has been pre-coated with an antibody specific to IL1RA. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to IL1RA. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain IL1RA, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of IL1RA in the samples is then determined by comparing the O.D. of the samples to the standard curve.

ncbi summary :
The protein encoded by this gene is a member of the interleukin 1 cytokine family. This protein inhibits the activities of interleukin 1, alpha (IL1A) and interleukin 1, beta (IL1B), and modulates a variety of interleukin 1 related immune and inflammatory responses. This gene and five other closely related cytokine genes form a gene cluster spanning approximately 400 kb on chromosome 2. A polymorphism of this gene is reported to be associated with increased risk of osteoporotic fractures and gastric cancer. Four alternatively spliced transcript variants encoding distinct isoforms have been reported. [provided by RefSeq, Jul 2008]
uniprot summary :
IL1RN: Inhibits the activity of interleukin-1 by binding to receptor IL1R1 and preventing its association with the coreceptor IL1RAP for signaling. Has no interleukin-1 like activity. Binds functional interleukin-1 receptor IL1R1 with greater affinity than decoy receptor IL1R2; however, the physiological relevance of the latter association is unsure. The intracellular form of IL1RN is predominantly expressed in epithelial cells. 4 isoforms of the human protein are produced by alternative splicing. Protein type: Vesicle; Cytokine; Secreted, signal peptide; Secreted. Chromosomal Location of Human Ortholog: 2q14.2. Cellular Component: extracellular space; cytoplasm; plasma membrane; intracellular. Molecular Function: interleukin-1 Type I receptor antagonist activity; interleukin-1 receptor binding; interleukin-1 Type II receptor antagonist activity; interleukin-1, Type II receptor binding; cytokine activity; interleukin-1, Type I receptor binding; interleukin-1 receptor antagonist activity. Biological Process: response to drug; positive regulation of JNK activity; negative regulation of heterotypic cell-cell adhesion; negative regulation of glutamate secretion; response to glucocorticoid stimulus; carboxylic acid metabolic process; response to lipopolysaccharide; female pregnancy; fever; sensory perception of pain; chronic inflammatory response to antigenic stimulus; memory; response to starvation; insulin secretion; acute-phase response; immune response; negative regulation of cytokine and chemokine mediated signaling pathway; lipid metabolic process; response to activity; negative regulation of membrane potential; negative regulation of cell migration; negative regulation of apoptosis.
