specificity :
This assay has high sensitivity and excellent specificity for detection of IL18BP. No significant cross-reactivity or interference between IL18BP and analogues was observed.
storage stability :
For unopened kit: All the reagents should be kept according to the labels on vials. The Standard, Detection Reagent A, Detection Reagent B and the 96-well strip plate should be stored at -20 degree C upon receipt while the others should be at 4 degree C. For opened kit: When the kit is opened, the remaining reagents still need to be stored according to the above storage condition. Besides, please return the unused wells to the foil pouch containing the desiccant pack, and reseal along entire edge of zip-seal. The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.
other info1 :
Samples: Mouse serum, plasma, tissue homogenates and other biological fluids. Assay Type: Sandwich. Detection Range: 62.5-4,000pg/mL. The standard curve concentrations used for the ELISA's were 4,000pg/mL, 2,000pg/mL, 1,000pg/mL, 500pg/mL, 250pg/mL, 125pg/mL, 62.5pg/mL. Sensitivity: The minimum detectable dose of IL18BP is typically less than 28.6pg/mL. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined by adding two standard deviations to the mean optical density value of twenty zero standard replicates and calculating the corresponding concentration.
other info2 :
Intended Uses: The kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of IL18BP in mouse serum, plasma, tissue homogenates and other biological fluids. Intra-assay Precision: Intra-assay Precision (Precision within an assay): 3 samples with low, middle and high level IL18BP were tested 20 times on one plate, respectively. Intra-Assay: CV<10%. Inter-assay Precision: Inter-assay Precision (Precision between assays): 3 samples with low, middle and high level IL18BP were tested on 3 different plates, 8 replicates in each plate. CV(%) = SD/meanX100. Inter-Assay: CV<12%
products description :
Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to IL18BP. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to IL18BP. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain IL18BP, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of IL18BP in the samples is then determined by comparing the O.D. of the samples to the standard curve.

ncbi summary :
The protein encoded by this gene functions as an inhibitor of the proinflammatory cytokine, IL18. It binds IL18, prevents the binding of IL18 to its receptor, and thus inhibits IL18-induced IFN-gamma production, resulting in reduced T-helper type 1 immune responses. This protein is constitutively expressed and secreted in mononuclear cells. Elevated level of this protein is detected in the intestinal tissues of patients with Crohn's disease. Alternatively spliced transcript variants encoding different isoforms have been described for this gene. [provided by RefSeq, Feb 2011]
uniprot summary :
IL18BP: Isoform A binds to IL-18 and inhibits its activity. Functions as an inhibitor of the early TH1 cytokine response. 3 isoforms of the human protein are produced by alternative splicing. Protein type: Secreted; Secreted, signal peptide. Chromosomal Location of Human Ortholog: 11q13. Cellular Component: extracellular space; extracellular region. Molecular Function: receptor antagonist activity; interleukin-18 binding. Biological Process: T-helper 1 type immune response; response to lipopolysaccharide
