The antibody recognizes all four isoforms of Hexokinase (1, 2) in recombinant protein. The antibody has been tested by ELISA and Western blot analysis to assure specificity and reactivity. Since application varies, however, each investigation should be titrated by the reagent to obtain optimal results. Recommended dilution range for Western blot analysis is 1:1,000 ~ 3,000. Recommended starting dilution is 1:2,000.

Hexokinase is the first enzyme in the glycolytic pathway, catalyzing the transfer of a phosphoryl group from ATP to glucose to form glucose-6-phosphate and ADP. In mammals, four distinct enzymes (types 1 to 4 hexokinases) have been identified. These enzymes are found in most cells, but there is tissue specificity for the particular type of hexokinase. Hexokinase 2 is found in the skeletal muscle and includes hydrophobic N-terminal sequence capable of targeting the hexokinase to mitochondria.

Hexokinases phosphorylate glucose to produce glucose-6-phosphate, the first step in most glucose metabolism pathways. This gene encodes hexokinase 2, the predominant form found in skeletal muscle. It localizes to the outer membrane of mitochondria.
