This assay has high sensitivity and excellent specificity for detection of ABCC8. No significant cross-reactivity or interference between ABCC8 and analogues was observed.
storage stability :
For unopened kit: All the reagents should be kept according to the labels on vials. The Standard, Detection Reagent A, Detection Reagent B and the 96-well strip plate should be stored at -20 degree C upon receipt while the others should be at 4 degree C. For opened kit: When the kit is opened, the remaining reagents still need to be stored according to the above storage condition. Besides, please return the unused wells to the foil pouch containing the desiccant pack, and reseal along entire edge of zip-seal. The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.
other info1 :
Samples: Human tissue homogenates, cell lysates and other biological fluids. Assay Type: Sandwich. Detection Range: 1.25-20ng/mL. The standard curve concentrations used for the ELISA's were 20ng/mL, 10ng/mL, 5ng/mL, 2.5ng/mL, 1.25ng/mL. Sensitivity: Typically less than 0.52ng/mL
other info2 :
Intra-assay Precision: Intra-assay Precision (Precision within an assay): 3 samples with low, middle and high level ABCC8 were tested 20 times on one plate, respectively. Intra-Assay: CV<10%. Inter-assay Precision: Inter-assay Precision (Precision between assays): 3 samples with low, middle and high level ABCC8 were tested on 3 different plates, 8 replicates in each plate. CV(%) = SD/meanX100. Inter-Assay: CV<12%
products description :
Intended Uses The kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of ABCC8 in human tissue homogenates, cell lysates and other biological fluids. Principle of the Assay: The microplate provided in this kit has been pre-coated with an antibody specific to ABCC8. Standards or samples are then added to the appropriate microplate wells with a biotin-conjugated antibody specific to ABCC8. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain ABCC8, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of ABCC8 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

ncbi summary :
The protein encoded by this gene is a member of the superfamily of ATP-binding cassette (ABC) transporters. ABC proteins transport various molecules across extra- and intra-cellular membranes. ABC genes are divided into seven distinct subfamilies (ABC1, MDR/TAP, MRP, ALD, OABP, GCN20, White). This protein is a member of the MRP subfamily which is involved in multi-drug resistance. This protein functions as a modulator of ATP-sensitive potassium channels and insulin release. Mutations and deficiencies in this protein have been observed in patients with hyperinsulinemic hypoglycemia of infancy, an autosomal recessive disorder of unregulated and high insulin secretion. Mutations have also been associated with non-insulin-dependent diabetes mellitus type II, an autosomal dominant disease of defective insulin secretion. Alternatively spliced transcript variants have been found for this gene. [provided by RefSeq, Dec 2013]
uniprot summary :
ABCC8: a multi-pass membrane protein subunit of the beta-cell ATP-sensitive potassium channel (KATP). Regulator of ATP-sensitive K(+) channels and insulin release. Associates with Kir6.2. Defects in ABCC8 are a cause of persistent hyperinsulinemic hypoglycemia of infancy (PHHI) [MIM:601820, 256450], and may contribute to non-insulin-dependent diabetes mellitus (NIDDM) in Northern European Caucasians. Two alternatively spliced human isoforms have been described. Protein type: Transporter; Membrane protein, integral; Transporter, ABC family; Membrane protein, multi-pass. Chromosomal Location of Human Ortholog: 11p15.1. Cellular Component: voltage-gated potassium channel complex; plasma membrane. Molecular Function: ATPase activity, coupled to transmembrane movement of substances; sulfonylurea receptor activity; potassium ion transmembrane transporter activity; ATP binding. Biological Process: synaptic transmission; carbohydrate metabolic process; energy reserve metabolic process; signal transduction; transmembrane transport; regulation of insulin secretion; potassium ion transport. Disease: Hyperinsulinemic Hypoglycemia, Familial, 1; Diabetes Mellitus, Transient Neonatal, 2; Diabetes Mellitus, Permanent Neonatal; Hypoglycemia, Leucine-induced; Diabetes Mellitus, Noninsulin-dependent
