Free radicals are highly reactive andgenerally short-lived species. Hence, moststudies of free radicals have beenrestricted to analysis of end productsformed following their interaction withcellular biomolecules. One technology thathas permitted direct study of free radicalsis electron spin resonance (ESR). For veryshort lived radicals, spin traps have beenemployed to generate longer lived radicalsthat are more amenable to ESR analysis.The most widely used of these spin trapsis 5,5-dimethyl-1-pyrroline-N-oxide(DMPO). The reaction of DMPO with aprotein radical gives rise to a nitroxideradical which can be disproportionate tohydrozylamine and nitrone adducts. Ofthese species, only the nitrone adduct canbe considered stable.
