specificity :
Osteonectin (SPARC) [Mouse monoclonal antibody to Osteonectin (SPARC)]. This antibody reacts with human and bovine osteonectin as determined by Western Blotting, and cross-reacts with rabbit and porcine osteonectin as determined by ELISA. This antibody reacts with both bone and platelet derived osteonectin; it does not react with non-activated platelets, but it does react with thrombin-stimulated platelets, as determined by Flow Cytometry. treatment with 5mM EDTA for 30 minutes, as determined by Flow Cytometry.
form :
100 ug IgG in 10 mM PBS, pH 7.4, lyophilized.
storage stability :
Lyophilized antibody is stable at 4 degree C for 2 years. Store the reconstituted solution in aliquots at -20 degree C for one year or at 4 degree C for 6 months. Avoid repeated freeze-thaw cycles.
tested application :
Western Blot, Flow Cytometry, Immunohistochemistry
app notes :
Western Blotting: 5-10 ug/ml, reducing and non-reducing conditions. Flow Cytometry: 1 ug/ml. Immunohistochemistry: 5-10 ug/ml, paraffin-embedded and frozen tissue sections.

ncbi summary :
Secreted protein acidic and rich in cysteine/osteonectin/BM40, or SPARC, is a matrix-associated protein that elicits changes in cell shape, inhibits cell-cycle progression, and influences the synthesis of extracellular matrix (ECM) (Bradshaw et al., 2003 [PubMed 12721366]).[supplied by OMIM]
uniprot summary :
Function: Appears to regulate cell growth through interactions with the extracellular matrix and cytokines. Binds calcium and copper, several types of collagen, albumin, thrombospondin, PDGF and cell membranes. There are two calcium binding sites; an acidic domain that binds 5 to 8 Ca2+ with a low affinity and an EF-hand loop that binds a Ca2+ ion with a high affinity. Subcellular location: Secreted extracellular space extracellular matrix basement membrane. Note: In or around the basement membrane. Ref.10 Ref.11 Ref.12 Ref.13. Developmental stage: Expressed at high levels in tissues undergoing morphogenesis, remodeling and wound repair. Sequence similarities: Belongs to the SPARC family.Contains 1 EF-hand domain.Contains 1 follistatin-like domain.Contains 1 Kazal-like domain. Sequence caution: The sequence AAA60993.1 differs from that shown. Reason: Contaminating sequence. Sequence of unknown origin in the C-terminal part.
